摘要
目的 探讨甲氨蝶呤和匹莫齐特联合使用对胃癌细胞系MKN45和MKN74的抑制效果。方法 培养人胃癌细胞系MKN45和MKN74,以每孔1.2×10^(4)个细胞的密度接种于96孔板中,每孔体积为100μL。配置相应浓度的MTX,使作用药物浓度分别为0.01μM、0.1μM、1μM、10μM和20μM;配置相应浓度的Pimozide,使作用药物浓度分别为0.1μM、0.5μM、1μM、2μM和4μM。设置空白对照孔:仅含培养基;细胞对照孔:仅含细胞和完全培养基;药物处理孔:含细胞和不同浓度药物(包括单独使用MTX、单独使用Pimozide以及MTX和Pimozide联合使用)。将96孔板置于二氧化碳孵箱中培养72 h后,使用CCK8检测细胞存活率,并使用CompuSyn软件计算药物协同效率。结果 单独使用0~1μM Pimozide对MKN45和MKN74细胞几乎没有抑制效果,2~4μM对MKN45和MKN74细胞有抑制效果(存活率<70%),且随着Pimozide浓度增加,抑制效果增加。单独使用MTX时,对MKN45和MKN74细胞均没有显著抑制作用。Pimozide和MTX联合使用后,当Pimozide浓度为4μM,MTX浓度为0.01~20μM时,MKN45和MKN74细胞活性受到十分显著抑制(细胞存活率<15%)。使用CompuSyn软件分析显示,Pimozide和MTX联合使用对MKN45和MKN74的抑制效果具有协同增效作用。结论 与单一药物相比,甲氨蝶呤和匹莫齐特联合使用可显著抑制胃癌细胞系MKN45和MKN74的活性,且具有协同增强效应。
Objective To investigate the inhibitory effect of methotrexate and pimozide on gastric cancer cell lines MKN45 and MKN74.Methods The human gastric cancer cell lines MKN45 and MKN74 were cultured and seeded in a 96-well plate at a density of 1.2×10^(4) cells per well with a volume of 100 μL per well.Configure the corresponding concentration of MTX so that the concentration of the effective drug is 0.01 μM,0.1 μM,1 μM,10 μM,and 20 μM;configure the corresponding concentration of Pimozide to make the concentration of the effective drug be 0.1 μM,0.5 μM,1 μM,2 μM,and 4 μM,respectively.Set blank control wells:only containing medium;cell control wells:containing only cells and complete medium;drug treatment wells:containing cells and different concentrations of drugs(including MTX alone,Pimozide alone,and combined use of MTX and Pimozide).After the 96-well plate was cultured in a carbon dioxide incubator for 72 hours,CCK8 was used to detect cell viability,and CompuSyn software was used to calculate the drug synergy efficiency.Results 0~1 μM Pimozide alone had almost no inhibitory effect on MKN45 and MKN74 cells,while 2~4μM had an inhibitory effect on MKN45 and MKN74 cells(survival rate <70%),and as the concentration of Pimozide increased,the inhibitory effect increased.When MTX is used alone,it has no significant inhibitory effect on MKN45 and MKN74 cells.After the combined use of Pimozide and MTX,when the concentration of Pimozide is 4 μM and the concentration of MTX is 0.01~20 μM,the cell viability of MKN45 and MKN74 is significantly inhibited(cell survival rate <15%).CompuSyn software was used to analyze the synergistic effect of Pimozide and MTX on the inhibitory effect of MKN45 and MKN74.Conclusion Compared with a single drug,the combined use of methotrexate and pimozide can significantly inhibit the activity of gastric cancer cell lines MKN45 and MKN74,and has a synergistic effect.
