摘要
以不同地理来源的华顶杜鹃叶片DNA为材料,采用PCR方法克隆叶绿体psbA-trnH,并利用生物信息学技术明确序列特征和系统发育关系。结果表明:华顶杜鹃psbA-trnH的序列长度为352~353 bp,GC值29.2%~29.3%;经多重比对后得出4个不同地理来源样品的psbA-trnH之间仅存在1个碱基的差异,但与其他杜鹃属植物同源序列间的差异较大;从系统发育分析得出,华顶杜鹃与其他14种杜鹃属植物在发育树上分为6组,其中,华顶杜鹃与长蕊杜鹃和毛棉杜鹃的亲缘关系最近,它们聚于同一分支。以期通过对华顶杜鹃叶绿体psbA-trnH序列的克隆与分析,为后续开展该珍稀濒危植物的遗传多样性研究奠定基础。
Chloroplast psbA-trnH was isolated from Rhododendron huadingense leaf DNA by PCR method,and bioinformatic techniques were applied to confirm its sequence characteristics and phylogenetic relationships.The results indicated that the sequence lengths varied from 352 to 353 bp,and their GC contents ranged from 29.2%to 29.3%.Multiple alignment results revealed there was only one base mutation among the sequences from four different geo‐graphical sources,however,larger differences were identified between psbA-trnH sequences from R.huadingense and other Rhododendron species.Phylogenetic analysis showed R.huadingense and other 14 Rhododendron plants were di‐vided into six groups,and R.huadingense,R.stamineum,and R.moulmainense shared the same clade,revealing their close relationship.Isolation and sequence analysis of chloroplast psbA-trnH shed an insight into future studies on genet‐ic diversity of this rare and endangered plant species.
