摘要
目的检测子痫前期(PE)患者胎盘组织中长链非编码RNA(lncRNA)生长停滞特异性转录本5(GAS5)表达水平,并探讨其临床意义。方法选取2018年6月至2020年5月在监利市人民医院行剖宫产的50例非重度PE患者(非重度PE组)、50例重度PE患者(重度PE组)及50例正常妊娠孕妇(对照组)作为研究对象。分娩后收集胎盘组织,检测胎盘组织螺旋动脉管壁厚度、螺旋动脉管腔面积;实时荧光定量聚合酶链反应法检测胎盘组织中lncRNA GAS5表达水平;Pearson法分析PE患者胎盘组织中lncRNA GAS5水平与螺旋动脉管壁厚度、螺旋动脉管腔面积的相关性。结果与对照组比较,非重度PE组、重度PE组胎盘组织螺旋动脉管壁厚度及lncRNA GAS5表达水平均升高,螺旋动脉管腔面积均降低,差异具有统计学意义(P<0.05);与非重度PE组比较,重度PE组胎盘组织螺旋动脉管壁厚度及lncRNA GAS5表达水平升高,螺旋动脉管腔面积降低,差异具有统计学意义(P<0.05)。PE患者胎盘组织中lncRNA GAS5表达水平与螺旋动脉管壁厚度存在正相关(r=0.485,P=0.000),与螺旋动脉管腔面积存在负相关(r=-0.614,P=0.000)。结论PE患者胎盘组织中lncRNA GAS5高表达,可能通过影响螺旋动脉灌注影响PE的发生发展,对其水平进行检测有助于临床判断PE患者病情。
Objective To detect the expression levels of long non-coding RNA(lncRNA)growth arrest specific transcript 5(GAS5)in placenta of patients with preeclampsia(PE)and to explore its clinical significance.Methods 50 cases of non-severe PE(non-severe PE group),50 cases of severe PE(severe PE group)and 50 cases of normal pregnant women(control group)undergoing cesarean section in Jianli People′s Hospital were selected,the placental tissue was collected after delivery,the wall thickness and lumen area of spiral artery were measured,the expression level of lncRNA GAS5 was detected by quantitative real-time polymerase chain reaction.Pearson method was used to analyze the correlation between lncRNA GAS5 level in placental tissue of PE and the wall thickness and lumen area of spiral artery.Results Compared with those in the control group,the wall thickness of spiral artery in placental tissue and the expression level of lncRNA GAS5 higher in non-severe PE group and severe PE group(P<0.05),and the lumen area of spiral artery lower(P<0.05),compared with those in non-severe PE group,the wall thickness of spiral artery in placental tissue and the expression level of lncRNA GAS5 were higher in severe PE group,and the lumen area of spiral artery lower(P<0.05).LncRNA GAS5 expression level was positively correlated with the wall thickness of spiral artery in the placenta of PE(r=0.485,P=0.000),and was negatively correlated with the lumen area of spiral artery(r=-0.614,P=0.000).Conclusions The expression of lncRNA GAS5 is high in placental tissue of PE patients,which may affect the occurrence and development of PE by affecting the perfusion of spiral artery.The detection of lncRNA GAS5 level is helpful for clinical judgment of PE.
