摘要
目的:观察温脾汤对脓毒症大鼠肠黏膜屏障功能的保护作用及其机制。方法:50只大鼠称重后按阿拉伯数字编号分为模型组、假手术组、中药低剂量组、中药中剂量组、中药高剂量组,每组各10只。中药低剂量组用温脾汤5.3g/(kg·d),中剂量组用温脾汤10.6g/(kg·d),高剂量组用温脾汤21.6g/(kg·d)。酶联免疫吸附测定法(ELISA)分析法测定血清D-乳酸、IL-17水平和肠道上皮细胞sIgA蛋白表达,备流式细胞实验检测Treg、Th17细胞,并进行组间比较分析。结果:相较于模型组,中药高剂量组、中剂量组、低剂量组血清D-乳酸、IL-17水平均更低(P<0.05)。相较于模型组,各组sIgA蛋白表达差异无统计学意义(P>0.05),但各用药组均有所降低。相较于模型组,中药低、中、高剂量组的Treg和Th17细胞含量均更高(P<0.05),且中药低、中、高剂量组之间呈明显上升趋势。结论:温脾汤对脓毒症大鼠肠黏膜屏障可起到保护作用,其机制可能与抑制炎症因子表达,调节免疫系统有关。
Objective:To observe the protective effect and mechanism of Warming Spleen Decoction on intestinal mucosal barrier function in sepsis rats.Methods:After weighing,50 rats were divided into model group,sham operation group,low-dose group of traditional Chinese medicine,middle-dose group of traditional Chinese medicine,and high-dose group of traditional Chinese medicine according to the Arabic numerals,with 10 rats in each group.The low-dose group of traditional Chinese medicine were given 5.3g/(kg·d)Warming Spleen Decoction,the middle-dose group given 10.6 g/(kg·d)Warming Spleen Decoction,and the high-dose group given 21.6 g/(kg·d)Warming Spleen Decoction.Enzyme-linked immunosorbent assay(ELISA)was used to determine serum D-lactic acid and IL-17 levels and the expression of sIgA protein in intestinal epithelial cells.Flow cytometry was used to detect Treg and Th17 cells,and the comparison between groups was analyzed.Results:Compared with the model group,the serum levels of D-lactic acid and IL-17 in the highdose,medium-dose,and low-dose groups of traditional Chinese medicine were lower(P<0.05).Compared with the model group,there was no significant difference in the expression of sIgA protein among the groups(P>0.05),but the drug groups were all decreased.Compared with the model group,the contents of Treg and Th17 cells in the low-dose,medium-dose and high-dose groups of traditional Chinese medicine were higher(P<0.05),and there was a significant upward trend among the low-dose,medium-dose and high-dose groups of traditional Chinese medicine.Conclusion:Warming Spleen Decoction can protect the intestinal mucosal barrier in sepsis rats,and its mechanism may be related to inhibiting the expression of inflammatory factors and regulating the immune system.
