拟南芥ABF3在逆境下响应的表达分析

摘要 本文以拟南芥ABF3基因编码区上游1631 bp的序列驱动的报告基因GUS的转基因拟南芥为对象,研究了ABF3基因的组织特异性、ABA处理、高盐处理、低渗处理等因素影响下的表达模式。基于启动子顺式作用元件库PLACE对ABF3上游序列的分析显示,该序列中含有多个与逆境应答、激素信号和光信号相关的顺式作用元件。q RT-PCR结果表明,在50 m M Na Cl处理下ABF3的m RNA含量无显著变化,而100和150 m M处理时,ABF3的m RNA含量升高至对照的3倍左右;50和100 m M山梨醇处理下,ABF3的m RNA含量分别为对照的1.7和2.4倍,处理浓度为150 m M时,ABF3的m RNA含量和对照无显著差异;在ABA处理下,ABF3的m RNA含量显著升高,10μM时ABF3的m RNA含量为对照的17倍。而低温处理下,ABF3的m RNA含量则显著降低,仅为对照的0.2倍。这些结果均表明ABF3在拟南芥苗期响应高盐、低温和ABA等多个过程。
