摘要
目的探讨用无水乙醇取代甲醇作淋巴细胞染色体、微核检测实验固定剂制片的效果优化。方法以剂量率为1Gy/min、剂量为2.5Gy的^(60)Coγ射线辐照正常非辐照接触者外周血,培养后,用无水乙醇∶冰醋酸=6∶1混合作为固定液,提纯,制得染色体分析片和微核片;与标准方法固定液无水甲醇∶冰醋酸=3∶1制片效果对比分析,将检测结果用JMP软件分别计算均值、t值和P值,并对所得数据用单项分析方法进行统计。结果镜下观察,无水乙醇替代实验和标准实验所制微核检测片效果均佳,无明显差异;替代实验所制染色体检测片较标准实验制片效果稍欠佳。微核分析结果对比,替代实验微核率均值为6.2‰,标准实验微核率均值为6.8‰,两组方法差异无统计学意义(t=1.236,P=0.232);染色体畸变分析结果对比,替代实验染色体畸变率均值为50.2%,标准实验染色体畸变率均值为51.2%,两组方法差异无统计学意义(t=1.213,P=0.241)。结论淋巴细胞染色体、微核检测实验中无水乙醇∶冰醋酸=6∶1作固定液所制检测片效果符合检测要求,在不影响结果和报告的准确性的同时,减少了对实验人员的危害,是值得推荐的试剂替代方法。
Objective To explore the optimization effect of using anhydrous ethanol instead of methanol as a fixative in tablet making for lymphocyte chromosome and micronucleus detection.Methods^(60)Coγ-ray at a dose rate of 1 Gy/min and a dose of 2.5 GY was used to irradiate the peripheral blood of normal non-irradiated contacts.After culture,the mixture(absolute ethanol∶glacial acetic acid=6∶1)was used as the fixed solution for purification.And then chromosome analysis tablets and micronucleus tablets were prepared.Compared with the standard method,the production effect of the fixed solution(anhydrous methanol∶glacial acetic acid=3∶1)was analyzed.The mean value,t value and P value were calculated by JMP software,and the obtained data were statistically analyzed by single analysis method.Results Microscopic observation showed that the effects of micronucleus detection tablets prepared by absolute ethanol substitute experiment and standard experiment were both good,and there was no significant difference.However,the effect of chromosome detection tablets prepared by substitute experiment was slightly inferior to tha of standard experiment.The results of micronucleus analysis showed that the mean micronucleus rate of alternative experiment was 6.2‰and that of standard experiment was 6.8‰.There was no significant difference between the two groups(t=1.236,P=0.232).The results of chromosome aberration analysis showed that the average chromosome aberration rate of substitute experiment was50.2%,while that of standard experiment was 51.2%.There was no significant difference between the two groups(t=1.213,P=0.241).Conclusions In the lymphocyte chromosome and micronucleus detection experiments,the effect of the test tablets prepared with the fixed solution(absolute ethanol∶glacial acetic acid=6∶1)met the detection requirements.It is a recommended reagen substitute method to reduce the harm to the experimenters without affecting the results and the accuracy of reports.
