摘要
为查明某猪场仔猪发病死亡原因,对该场送检的2头发病仔猪进行病理剖检,无菌采集肺脏、脾脏、心脏等组织样品,进行细菌分离鉴定及常发病毒病猪繁殖与呼吸综合征病毒(PRRSV)、猪圆环病毒2型(PCV2)、猪伪狂犬病病毒(PRV)、猪瘟病毒(CSFV)、猪流感病毒(SIV)、猪细小病毒(PPV)等病原的PCR检测,并对核酸阳性的病原进行测序和遗传进化分析。结果表明,病料中未分离到致病菌;PCR检测PRRSV和PCV2呈阳性。对PRRSV的ORF5基因和PCV2的ORF2基因进行遗传进化分析发现,检测到的PRRSV属于NADC30-like亚群,PCV2属于PCV-2b基因型。通过病理剖检和实验室检测,确定该猪场存在PRRSV和PCV2的混合感染,建议该场采取消毒隔离、紧急免疫、防止继发感染等措施,成功控制了病情的发展,研究结果为猪场相关疾病的诊断及防控提供了一定的参考。
In order to identify the cause of morbidity and death of piglets in a pig farm,pathological autopsy was performed in two sick piglets sent for detection,and tissue samples of lung,spleen and heart were collected aseptically for bacterial isolation and identification,and PCR testing of pathogens such as porcine reproductive and respiratory syndrome virus(PRRSV),porcine circovirus type 2(PCV2),porcine pseudorabies virus(PRV),classical swine fever virus(CSFV),swine influenza virus(SIV)and porcine parvovirus(PPV)for frequent viral diseases,and sequencing and genetic evolutionary analysis were performed in the nucleic acid-positive pathogens.The results showed that no pathogenic bacteria were isolated from the diseased materials;PCR was positive for PRRSV and PCV2.Genetic evolutionary analysis of the ORF5 gene of PRRSV and the ORF2 gene of PCV2 revealed that the detected PRRSV belonged to the NADC30-like subgroup and PCV2 belonged to the PCV2b genotype.Through pathological dissection and laboratory tests,it was determined that there was a mixed infection of PRRSV and PCV2 in the farm,and the farm was advised to take measures such as disinfection and isolation,emergency immunization and prevention of secondary infection,which successfully controlled the development of the disease,and the results of the study provided some references for the diagnosis,prevention and control of related diseases in the farm.
