摘要
为了研究金黄色葡萄球菌对奶牛乳房炎的致病机制,以及为后续药物预防与治疗的深入研究,建立由临床奶汁中分离纯化的金黄色葡萄球菌引起的大鼠乳腺炎模型。试验分为对照组和试验组共6组,每组6只,试验组金黄色葡萄球菌分为2×10^(5)、2×10^(6)、2×10^(7)、2×10^(8)、2×10^(9)CFU/mL共5个细菌含量浓度,制备好的细菌悬液经大鼠第4对乳头导管注菌,每侧100μL。研究表明,从患乳房炎的奶牛乳汁中分离纯化出金黄色葡萄球菌,临床表现及乳腺组织眼观变化,随着金黄色葡萄球菌数量增多,组织病变逐渐加重,且M5组死亡2只;病理组织切片发现,与对照组相比,M1、M2组炎症较轻,腺泡腔只出现少量炎性细胞浸润,M3、M4组腺泡间隙出现明显的渗出液,腺泡腔内明显可见一定数量的炎性细胞浸润,M5组组织破坏严重,腺泡腔出现大量炎性细胞浸润;组织细菌计数结果与注菌浓度成正比;血常规结果发现,与对照组相比,白细胞、中性粒细胞及血小板数量都在M3组开始出现显著或极显著升高;组织中IL-1β、IL-6、TNF-α表达水平都随注菌数量增多而升高,与对照组相比,IL-1β、TNF-α在M3组开始出现显著或极显著升高,IL-6含量在M2组出现极显著增高;TLR2及p-p65蛋白表达,分别在M3组和M4组开始出现极显著升高。成功建立大鼠乳腺炎模型,金黄色葡萄球菌引起乳腺炎的最佳含菌量为2×10^(8)CFU/mL。
In order to better study the pathogenic mechanism of Staphylococcus aureus on mastitis in dairy cows,as well as for the subsequent in-depth study of drug prevention and treatment,a rat mastitis model caused by Staphylococcus aureus isolated and purified from clinical milk was established.The test was divided into six groups of six animals each for the control and test groups.Five concentrations of 2×10^(5),2×10^(6),2×10^(7),2×10^(8),2×10^(9)CFU/mL were set for the Staphylococcus aureus group.Microinjector was used to inject the bacterial suspension into the fourth pair of mammary glands of rats,with 100μL per side.Research showed successful isolation and purification of Staphylococcus aureus from the milk of cows with mastitis.As the concentration of injected Staphylococcus aureus got higher and higher,the mammary gland tissue lesions gradually worsen and two deaths occurred in the M5 group.The pathological section showed that compared with the control group,the inflammation in M1 and M2 groups was mild and only a small amount of inflammatory cell infiltration in the lumen of the glandular follicle.M3 and M4 groups showed exudate between the mammary follicles and a certain number of inflammatory cells infiltrated in the lumen of the follicles were clearly visible.Group M5 had severe destruction of mammary tissue and massive inflammatory cell infiltration in the alveolar cavity.Bacterial count results in mammary tissue were proportional to the concentration of injected Staphylococcus aureus.Routine blood results revealed that compared with the control group,white blood cell,neutrophil and platelet numbers all started to show significant or highly significant increases or decreases in the M3 group;The expression levels of IL-1β,IL-6,and TNF-αin mammary tissue all increased with the higher concentration of injected bacteria.Compared with the control group,IL-1βand TNF-αstarted to show significant or highly significant increases in the M3 group,and IL-6 showed highly significant increases in the M2 group.TL
