摘要
目的 运用网络药理学方法探讨五味消毒饮治疗痛风性关节炎(GA)的作用机制。方法 通过检索各类数据库和运用相关软件,确定五味消毒饮与GA的交集基因;构建“化合物-交集基因-疾病”网络模型并确定核心基因;运用R语言进行基因本体功能富集分析和京都基因与基因组百科全书通路注释富集分析。结果 五味消毒饮与GA的交集基因共19个;GO功能富集中细胞因子受体结合、受体配体活性、细胞因子活性为主要的生物过程;KEGG通路注释富集中类风湿性关节炎通路、NOD样受体信号通路等为主要通路。结论 通过对五味消毒饮治疗GA作用机制的研究,揭示五味消毒饮多成分、多靶点、多途径的作用特点。
OBJECTIVE To explore the mechanism of Wuwei Xiaodu drink in the treatment of gouty arthritis(GA) by network pharmacology.METHODS The intersection genes of Wuwei Xiaodu drink and GA were screened by searching different databases and softwares.Cytoscape software was used to construct the network model of "compound-intersection genes-disease",and the hub genes were identified.GO function and KEGG pathway annotation was performed by R software.RESULTS A total of 19 genes were identified for the intersection of Wuwei Xiaodu drink and GA.cytokine receptor binding and receptor ligand activity were the main GO biological processes.Rheumatoid arthritis and NOD-like receptor signaling pathway were the main pathways of KEGG.CONCLUSION Through the study on the mechanism of Wuwei Xiaodu drink treating GA,the characteristics of the the traditional Chinese medicine prescription treating with multi-component,multi-target and multi-pathway are revealed.
