miRNA-146b-5p靶向调节LAD-1对非小细胞肺癌增殖和凋亡的影响 被引量:3

摘要 目的探讨miRNA-146b-5p靶向调节线皮素-1(LAD-1)对非小细胞肺癌(NSCLC)增殖和凋亡的影响。方法选取我院2018年3月至2021年1月接收的45例诊断为NSCLC并进行肿瘤根治术的患者,留存肿瘤组织及癌旁组织作为检测样本。采用RT-PCR检测样本中miRNA-146b-5p相对表达量。复苏NSCLC A549细胞,并建立稳定A549敲降miRNA-146b-5p细胞及过表达miRNA-146b-5p细胞,依次设定为对照组、敲降组及过表达组。采用CCK-8法、流式细胞术及蛋白印迹法分别测定三组的细胞增殖率、凋亡率及LAD-1蛋白相对表达量。结果肿瘤组织中miRNA-146b-5p相对表达量明显高于癌旁组织,差异具有统计学意义(P<0.05)。敲降组的miRNA-146b-5p相对表达量明显低于对照组(P<0.05);过表达组的miRNA-146b-5p相对表达量明显高于对照组(P<0.05)。培养12、24、48、72 h,敲降组的细胞增殖率低于对照组,过表达组的细胞增殖率高于对照组,差异具有统计学意义(P<0.05)。敲降组的细胞凋亡率高于对照组(P<0.05);过表达组的细胞凋亡率低于对照组(P<0.05)。敲降组的LAD-1蛋白相对表达量明显低于对照组(P<0.05);过表达组的LAD-1蛋白相对表达量明显高于对照组,差异具有统计学意义(P<0.05)。结论miRNA-146b-5p在NSCLC肿瘤组织中呈高表达,其可能通过显著上调LAD-1蛋白表达来促使NSCLC肿瘤细胞增殖并抑制凋亡,进而促进癌症发展。 Objective To investigate the effects of miRNA-146b-5p targeting regulation of ladinin-1(LAD-1)on proliferation and apoptosis of non-small cell lung cancer(NSCLC).Methods Forty-five patients with NSCLC who underwent radical resection in our hospital from March 2018 to January 2021 were selected.Tumor tissues and adjacent tissues were retained as test samples.The relative expression of miRNA-146b-5p in the samples was detected by RT-PCR.NSCLC A549 cells were revived,and stable A549 knock-down miRNA-146b-5p cells and overexpression miRNA-146b-5p cells were established,which were set as control group,knock-down group and overexpression group in turn.The cell proliferation rate,apoptosis rate and the relative expression of LAD-1 protein in the three groups were determined by CCK-8 method,flow cytometry and Western Blotting respectively.Results The relative expression of miRNA-146b-5p in tumor tissues was significantly higher than that in adjacent tissues,and the difference was statistically significant(P<0.05).The relative expression of miRNA-146b-5p in the knock-down group was significantly lower than that in the control group(P<0.05);the relative expression of miRNA-146b-5p in the overexpression group was significantly higher than that in the control group(P<0.05).After 12,24,48 and 72 h of culture,the cell proliferation rate in the knock-down group was lower than that in the control group,the cell proliferation rate in the overexpression group was higher than that in the control group,and the differences were statistically significant(P<0.05).The apoptosis rate in the knock-down group was higher than that in the control group(P<0.05);the apoptosis rate in the overexpression group was lower than that in the control group(P<0.05).The relative expression of LAD-1 protein in the knock-down group was significantly lower than that in the control group(P<0.05);the relative expression of LAD-1 protein in the overexpression group was significantly higher than that in the control group,and the difference was statistically si
