酸枣仁汤对老年慢性睡眠剥夺大鼠节律基因与钟控基因表达的影响 被引量:7

Effects of Suanzaoren Decoction on the expression of rhythm gene and clock control gene in elderly chronic sleep deprivation rats

摘要 目的:研究酸枣仁汤对老年慢性睡眠剥夺模型大鼠节律基因与钟控基因表达的影响。方法:将50只雄性Wistar大鼠随机分为空白对照组,模型对照组,阳性对照组(0.18 mg·kg^(-1)·d^(-1)),酸枣仁汤高、低剂量组(12.96、6.48 g·kg^(-1)·d^(-1)),除空白对照组外,其余各组均皮下注射D-半乳糖,各组大鼠按0.125 g·kg^(-1)·d^(-1)剂量给药,给药容量为0.5 mL/100 g,每日1次,连续42 d,于末次给药结束后进行多平台水环境慢性睡眠剥夺,每天剥夺18 h,连续剥夺21 d,制作老年慢性快动眼睡眠剥夺模型,造模结束后各组开始灌胃给药,给药容量均为2 mL/100 g,每日1次,连续7 d,空白对照组、模型对照组给予等体积的0.9%氯化钠溶液。采用自主活动仪检测酸枣仁汤对老年慢性快动眼睡眠剥夺模型大鼠自主活动的影响,采用Real-Time PCR和免疫组化法分别检测各组大鼠下丘脑生物钟基因Clock、Bmal1、Per1、Cry1和钟控基因Rev-erbα、RorαmRNA和蛋白表达水平。结果:与空白对照组比较,模型对照组大鼠光照环境活动路程、黑暗环境活动路程、总活动路程均显著增加(P<0.01),Clock、Bmal1、Per1、Cry1、Rev-erbα、Rorα的mRNA和蛋白表达水平显著降低(P<0.01);与模型对照组比较,阳性对照组、酸枣仁汤高剂量组光照环境活动路程、黑暗环境路程、总活动路程均显著减少(P<0.01),Clock、Bmal、Per1、Cry1、Rev-erbα、Rorα的mRNA和蛋白表达水平显著升高(P<0.01,P<0.05);酸枣仁汤低剂量组Bmal1、RorαmRNA表达显著升高(P<0.01,P<0.05)。结论:酸枣仁汤可以改善大鼠自主活动,其机制可能与上调节律基因Clock、Bmal1、Per1、Cry1与钟控基因Rev-erbα、Rorα的表达有关。 Objective:To study the effects of Suanzaoren Decoction on the expression of rhythm gene and clock control gene in elderly rats with chronic sleep deprivation.Methods:Fifty male Wistar rats were randomly divided into blank control group,model control group,positive control group(0.18 mg·kg^(-1)·d^(-1)),high and low dose Suanzaoren Decoction groups(12.96,6.48 g·kg^(-1)·d^(-1)).Except the blank control group,the other groups were subcutaneously injected with D-galactose.The rats in each group were administered at the dose of 0.125 g·kg^(-1)·d^(-1),with the dosage of 0.5 mL/100 g once a day for 42 days,After the last administration,chronic sleep deprivation in multi platform water environment was carried out for 18 hours a day and 21consecutive days.The elderly chronic rapid eye movement sleep deprivation model was made.After the modeling,each group began to give drugs by gavage,with the dosage of 2 mL/100 g,once a day for 7 consecutive days,the blank control group and model control group were given equal volume of 0.9%sodium chloride solution.The effect of Suanzaoren Decoction on the autonomic activity of elderly rats with chronic REM sleep deprivation model was detected by autonomic activity instrument.The hypothalamic clock genes Clock,Bmal1,Per1,Cry1 and clock controlled gene Rev-erbα,Rorαwere detected by real time PCR and immunohistochemistry.Results:Compared with the blank control group,the activity distance and time in the light environment,the activity distance and time in the dark environment,the total activity distance and time in the model control group were significantly increased(P<0.01),the mRNA and protein expression levels of Clock,Bmal1,Per1,Cry1 and Rev-erbα,Rorαdecreased significantly(P<0.01).Compared with the model control group,the activity distance and time in the light environment,the distance and time in the dark environment,the total activity distance and time in the positive control group and the high dose Suanzaoren Decoction group were significantly reduced(P<0.01),the mRNA and p
