摘要
目的观察5个月规律不同强度有氧运动对APP/PS1双转基因小鼠海马齿状回双皮质素及β-淀粉样蛋白的影响。方法将3月龄APP/PS1双转基因小鼠随机分为5组:空白对照组(AD^(C))、低强度运动组(AD^(L))、中强度运动组(AD^(M))、大强度运动组(AD^(H)),另设野生型组(WT^(C)),每组8只。运动组以不同强度有氧运动进行干预:低强度跑速为12 m/min,中强度跑速为15 m/min,大强度跑速为18 m/min。每周5 d,每天30 min,持续5个月。在各运动强度下,当小鼠停止不运动时,不对小鼠进行电刺激,只轻柔地触摸尾巴让小鼠继续运动。然后,通过Morris水迷宫及八臂迷宫实验评估小鼠的学习和记忆能力,利用免疫荧光和Western blot检测小鼠海马双皮质素及Aβ_(1-42)的蛋白表达水平。结果(1)在MWM定位航行实验过程中,各组小鼠的逃避潜伏期都是逐渐减少的。从第2天开始,AD^(C)组小鼠的平均逃避潜伏期就显著高于WT^(C)组(P<0.05);与AD^(C)组及AD^(L)组相比,AD^(M)组小鼠从第3天开始显著降低,AD^(H)组小鼠从第4天开始显著降低(P<0.05)。各组小鼠之间的游泳速度无显著差异(P>0.05)。在MWM定位航行实验中,与WT^(C)组小鼠相比,AD^(C)组穿越原平台所在区域的次数显著降低(P<0.01);与AD^(C)组及AD^(L)组相比,AD^(M)组、AD^(H)组小鼠穿越原平台所在区域的次数显著升高(P<0.01,P<0.05)。(2)就工作记忆错误而言,AD^(C)组小鼠错误次数显著高于WT^(C)组(P<0.01);AD^(M)组、AD^(H)组小鼠错误次数显著低于AD^(C)组(P<0.01);AD^(M)组、AD^(H)组小鼠错误次数显著低于AD^(L)组(P<0.01)。就参考记忆错误而言,AD^(M)组小鼠错误次数显著低于AD^(C)组(P<0.05);AD^(M)组、AD^(H)组小鼠错误次数显著低于AD^(L)组(P<0.05)。(3)与WT^(C)组相比,AD^(C)组海马DCX表达较低,而Aβ_(1-42)表达较高(P<0.05);与AD^(C)组相比,AD^(M)、AD^(H)组DCX表达显著提高,而Aβ_(1-42)表达显著降低(P<0.05,P<0.01);与AD^(L)组相比,AD^(M)
Objective To investigate the effect of 5 months aerobic exercise of different intensity on doublecortin andβ-amyloid protein in dentate gyrus of hippocampus of 8-month-old APP/PS1 double transgenic mice.Methods 3-month-old APP/PS1 dual-transgenic AD mice were randomly divided into five groups.Each group has 8 mice,and they are Control group(AD^(C)),Low-loaded exercise group(AD^(L)),Medium-loaded exercise group(AD^(M))and High-loaded exercise group(AD^(H)),as well as Wild-type control group(WT^(C)).AD^(C) and WT^(C) group mice were fed naturally for 5 months.Each exercise group was treated with aerobic exercise of different intensities:Low-loaded was 12 m/min running speed,medium-loaded 15 m/min,and high-loaded 18 m/min.5 days a week for 30 minutes a day for 5 months.At each exercise intensity,when the mice stopped moving,no electrical stimulation was given to the mice,only gentle touch of the tail to let the mice continue to move.Then,the Morris Water Maze(MWM)test and the 8-arm radial maze test were performed to estimate mice’learning and memory abilities,while the immunofluorescence and Western blot technique were used to determine the expression levels of DCX and Aβ_(1-42) in the hippocampus.Results(1)In the process of navigation training,all mice’escape latencies gradually decreased.From the second day,the average escape latency of the AD^(C) group was significantly higher than that of the WT^(C) group(P<0.05).Compared with the AD^(C) group and the AD^(L) group,escape latency in the AD^(M) group decreased significantly from the third day,and that in the AD^(H) group decreased significantly from the fourth day(P<0.05).There was no significant difference in swimming speed between these groups(P>0.05).In the MWM navigation experiment,for the times of crossing the original platform area,AD^(C) group was significantly lower than WT^(C) group(P<0.01),AD^(M) group and AD^(H) group was significantly higher than AD^(C) group and AD^(L) group(P<0.01,P<0.05).(2)In terms of working memory errors,the number of error
