摘要
为了建立猪繁殖与呼吸综合征病毒-2(PRRSV-2)抗体检测方法,试验采用原核表达猪繁殖与呼吸综合征病毒NSP7蛋白,并以该蛋白为基础建立间接ELISA方法,对该方法的特异性、重复性、敏感性以及与爱德士商品化试剂盒符合率进行比较。试验结果表明,PRRSV-2 NSP7蛋白表达良好,可获得可溶蛋白,建立NSP7间接ELISA方法特异性、重复性、敏感性良好,与爱德士商品化试剂盒的符合率96.2%,说明PRRSV-2 NSP7间接ELISA检测方法建立成功。
In order to establish a new detection method for PRRSV-2 antibody,the prokaryotic expression was used to express PRRSV-2 NSP7 protein.Based on the NSP7 protein,an indirect ELISA method was established,and the specificity,repeatability,and the cross reactivity and the similarity with IDEXX detection kit were studied.The results showed that the NSP7 protein of PRRSV-2 was well expressed and the soluble protein could be obtained.Based on this,the indirect ELISA method was established,and the similarity with IDEXX detection kit was 96.2%.The detection results of clinical samples showed that it could be used to detect clinical samples with good specificity and repeatability.The indirect ELISA method for NSP7 protein of porcine reproductive and respiratory syndrome virus was successfully established.
