摘要
目的探讨卡维地洛对溃疡性结肠炎(UC)模型大鼠脂质过氧化产物和炎症因子的影响。方法30只大鼠随机分成5组①空白对照组(Cont);②乙酸处理组(AA);③卡维地洛预处理组(CarV),使用卡维地洛30 mg·kg^(-1)1次·d^(-1)灌胃预处理;④卡维地洛治疗组(CarV+AA),使用卡维地洛30 mg·kg^(-1)1次·d^(-1)灌胃对UC大鼠干预;⑤美沙拉嗪治疗组(MES+AA),使用美沙拉嗪100 mg·kg^(-1)1次·d^(-1)灌胃对UC大鼠干预,每组6只。应用乙酸诱导UC大鼠模型,建构UC大鼠模型后,所有治疗组均治疗7 d,其他组使用等量盐水替代。检测各组大鼠结肠组织湿重、黏液含量、组织病理学改变及脂质过氧化物(LPO)代谢产物丙二醛(MDA)、炎症因子、PGE_(2)、NO水平变化。结果AA组大鼠结肠组织中MDA和炎症因子、PGE_(2)、NO水平、结肠湿重均明显升高(P<0.05或P<0.01),黏液含量降低(P<0.01)。CarV+AA组大鼠结肠组织中炎症因子、PGE_(2)、NO水平、结肠湿重较AA组大鼠明显降低(P<0.05或P<0.01),黏液含量增多(P<0.01);MDA水平降低,但和美沙拉嗪组比较差异仍有统计学意义(P<0.01),与AA组比较差异无统计学意义(P>0.05)。结论卡维地洛可降低乙酸诱导的UC模型大鼠肠黏膜损伤,可能与其抗炎作用和抗氧化特性相关。
Objective To investigate the effect of carvedilol on lipid peroxidation products and inflammatory factors in a rat model of ulcerative colitis.Methods A total of 30 rats were randomly divided into 5 groups:①blank control group(Cont);②acetic acid treatment group(AA);③carvedilol pretreatment group(CarV),treated with carvedilol 30 mg·kg^(-1)1·d^(-1) gavage pretreatment;④Carvedilol treatment group(CarV+AA),UC rats were treated with carvedilol 30 mg·kg^(-1)1·d^(-1) gavage;⑤In the mesalazine treatment group(MES+AA),UC rats were intervened with mesalazine 100 mg·kg^(-1)1·d^(-1) by gavage.There were 6 rats in each group.The UC rat model was induced by acetic acid.After the UC rat model was established,all treatment groups were treated for 7 days,and the other groups were replaced with the same amount of saline.The wet weight,mucus content,lipid peroxide(LPO)metabolite malondialdehyde(MDA),histopathological changes,and inflammatory factors,PGE_(2),and NO levels in the colon tissue of rats in each group were detected.Results The levels of MDA and inflammatory factors,PGE_(2),NO,and colonic wet weight in colon tissue of rats in AA group were significantly increased(P<0.05 or P<0.01),and the content of mucus was decreased(P<0.01).Compared with the AA group,the inflammatory factors,PGE_(2),NO levels and colon wet weight in the colon tissue of the CarV+AA group were significantly lower(P<0.05 or P<0.01),and the mucus content was increased(P<0.01);however,compared with the mesalazine group,the difference was still statistically significant(P<0.01),and there was no significant difference compared with the AA group(P>0.05).Conclusion Carvedilol can reduce the intestinal mucosal injury induced by acetic acid in a rat model of ulcerative colitis,which may be related to its anti-inflammatory and antioxidant properties.
