摘要
目的:同时测定芍药苷和甘草酸单铵盐的含量,初步探索自制芍药甘草汤有效部位的质量控制方法,为下一步进行有效部位的药效学以及质量控制的深入研究奠定基础。方法:采用反相高效液相色谱法测定芍药苷和甘草酸单铵盐的含量,色谱柱为ZORBAX SB-C18柱(150 mm×2.1 mm,1.8μm),以甲醇-0.4%甲酸溶液(体积比15∶85~45∶55)为流动相进行梯度洗脱,检测波长为238 nm,柱温为30℃。结果:芍药苷的线性范围为0.0406~0.6090 mg·mL^(-1),甘草酸单铵盐的线性范围为0.0437~0.6550 mg·mL^(-1),芍药苷的平均回收率为99.38%,甘草酸单铵盐的平均回收率为99.93%。结论:该方法能准确快速地进行芍药甘草汤有效部位中芍药苷和甘草酸单铵盐的含量测定,并为白芍-甘草药对的研发奠定基础。
Objective To determine paeoniflorin and glycyrrhizic acid in effective fraction from Shaoyao Gancao decoction at the same time,the quality control method of effective fraction from self-made Shaoyao Gancao decoction was preliminarily explored,and to lay the foundation for further research of pharmacodynamics and quality control of effective parts.Methods The determination was performed by RP-HPLC using ZORBAX SB-C18 column(150 mm×2.1 mm,1.8μm),methanol-0.4% methanoic acid solution(15∶85~45∶55)as the mobile phase for gradient elution analysis,the wavelength was 238 nm and column temperature was 30℃.Results The linearity of paeoniflorin was 0.0406~0.6090 mg·mL^(-1) and 0.0437~0.6550 mg·mL^(-1) for glycyrrhizic acid.The average recovery was 99.38% and 99.93%respectively.Conclusion This method is simple rapid and accurate for the quality control of the effective fraction from Shaoyao Gancao decoction,and lay a foundation for the research and development of Radix paeoniae rubra-Radix et rhizoma glycyrrhizae Pair.
