实验性牙周炎大鼠牙周组织中趋化因子受体CCR5、CXCL12、RANTES表达及意义 被引量:2

摘要 目的:探讨实验性牙周炎大鼠牙周组织中趋化因子受体5(CCR5)、C-X-C基趋化因子12(CXCL12)、T细胞表达和分泌因子(RANTES)表达水平及意义。方法:将100只SD大鼠分成正常对照组与4周、8周、12周、16周牙周炎建模治疗组。建模成功后,各治疗组给予相应措施治疗,并分别于相应时间点处死,获取牙周组织进行牙槽骨吸收(腭侧多边形面积)测定,RT-PCR及蛋白印迹法测定牙周组织CCR5、CXCL12、RANTES mRNA与蛋白表达水平。结果:与正常对照组比较,4周、8周、12周、16周建模治疗组腭侧多边形面积明显升高(P<0.05),且随建模时间延长,腭侧多边形面积逐渐降低(P<0.05)。与正常对照组比较,4周、8周、12周、16周建模治疗组牙周组织CCR5、CXCL12、RANTES mRNA和蛋白水平明显升高(P<0.05),且随建模时间延长,CCR5、CXCL12、RANTES mRNA和蛋白水平逐渐降低,差异有统计学意义(P<0.05)。结论:实验性牙周炎大鼠牙周组织中CCR5、CXCL12、RANTES mRNA与蛋白表达水平升高,其与牙周炎症反应密切相关。 Objective:To investigate the expression of chemokines CCR5,CXCL12 and RANTES in the periodontal tissues of experimental periodontitis rats and its significance.Methods:One hundred SD rats were divided into: normal control group, 4 weeks, 8 weeks, 12 weeks, 16 weeks periodontitis modeling treatment group.After successful modeling, the treatment groups were given corresponding treatments and sacrificed at corresponding time points.Periodontal tissues were obtained for the determination of alveolar bone resorption(palatal polygonal area),and the CCR5,CXCL12,RANTES mRNA and protein expression levels in periodontal tissue were determined by RT-PCR and Western blotting respectively.Results:Compared with the normal control group, the area of the palatal polygonal polygon in the modeling treatment group of 4 weeks, 8 weeks, 12 weeks, and 16 weeks was significantly increased(P<0.05),and with the extension of the modeling treatment group, the area of the palatal polygonal polygon gradually decreased(P<0.05).Compared with the normal control group, the CCR5,CXCL12,RANTES mRNA and protein levels in the periodontal tissues of the 4 weeks, 8 weeks, 12 weeks, and 16 weeks modeling treatment group were significantly increased(P<0.05),and with the extension of the modeling treatment group, in periodontal tissues gradually decreased(P<0.05).Conclusion:The expression levels of chemokines CCR5,CXCL12,RANTES mRNA and protein in the periodontal tissues of experimental periodontitis rats are increased, which are closely related to periodontal inflammation.
