舒芬太尼对肾缺血再灌注大鼠肾组织细胞自噬的影响

Effects of sufentanil on autophagy of renal tissue cells in rats with renal ischemia reperfusion injury

摘要 目的:探讨舒芬太尼(Sufen)对肾缺血再灌注(RIRI)大鼠肾组织细胞自噬的影响及其作用机制。方法:将60只雄性SD大鼠随机分为对照组、模型组、Sufen组、LY294002组、Sufen+LY294002组,每组12只,除对照组外,其他组均需构建RIRI模型。检测大鼠血清中肌酐(Cr)、血尿素氮(BUN)、丙二醛(MDA)和超氧化物歧化酶(SOD)活性;Western blotting检测大鼠肾脏组织中微管相关蛋白1轻链3(LC3)、p62、磷脂酰肌醇-3-激酶(PI3K)、PI3K、蛋白激酶B(AKT)、p-AKT蛋白表达。结果:与模型组比较,Sufen组大鼠血清中Cr、BUN水平降低,肾脏组织中的肾小管损伤减轻,肾脏组织中MDA含量、自噬小体的数量、LC3-Ⅱ/LC3-Ⅰ降低,SOD活性、p62蛋白、p-PI3K/PI3K、p-AKT/AKT水平升高,而LY294002组上述对应指标与Sufen组呈相反趋势(均P<0.05);与Sufen组比较,Sufen+LY294002组大鼠血清中Cr、BUN水平升高,肾脏组织中的肾小管损伤严重,肾脏组织中MDA含量、自噬小体的数量、LC3-Ⅱ/LC3-Ⅰ升高,SOD活性、p62蛋白、p-PI3K/PI3K、p-AKT/AKT水平降低(均P<0.05)。结论:舒芬太尼可能通过激活PI3K/AKT通路来抑制过度自噬,进而发挥对RIRI大鼠肾脏的保护作用。 Objective:To investigate the effects of sufentanil(Sufen)on the autophagy of renal tissue cells in rats with renal ischemia reperfusion injury(RIRI)and its mechanism.Methods:Sixty male SD rats were randomly divided into control group,model group,Sufen group,LY294002 group and Sufen+LY294002 group,12 rats in each group.RIRI model was established in all groups except for the control group.Creatinine(Cr),blood urea nitrogen(BUN),malondialdehyde(MDA)and superoxide dismutase(SOD)activities were detected in rat serum.The expressions of microtubule-associated protein 1 light chain 3(LC3),p62,phosphatidylinositol-3-kinase(PI3K),p-PI3K,protein kinase B(AKT)and p-AKT in rat kidney tissue were detected by western blotting.Results:Compared with the model group,the serum levels of Cr and BUN,tubular damage in renal tissues,MDA content,number of autophagic vesicles and LC3-Ⅱ/LC3-Ⅰin renal tissues were reduced in the Sufen group,whereas the SOD activity,p62 protein level,and p-PI3K/PI3K and p-AKT/AKT ratios were increased,while the corresponding indexes in the LY294002 group showed the opposite trend to those in the Sufen group(P<0.05).Compared with the Sufen group,the rats in the Sufen+LY294002 group had increased serum levels of Cr and BUN,severe tubular damage in renal tissues,increased MDA content,number of autophagic vesicles and LC3-Ⅱ/LC3-Ⅰ,along with decreased SOD activity,p62 protein level,p-PI3K/PI3K and p-AKT/AKT in renal tissues(P<0.05).Conclusion:Sufentanil may inhibit excessive autophagy by activating the PI3K/AKT pathway,which in turn exerts a protective effect on the kidney of RIRI rats.
