SDF-1α/CXCR4生物轴促进静脉移植HUCMSCs在MCAO小鼠体内的迁移和功能恢复 被引量:1

摘要 目的探讨静脉移植HUCMSCs通过SDF-1α/CXCR4生物轴参与改善脑缺血的作用机制,明确中药通心络胶囊干预后的影响。方法采用随机法将小鼠分组,共4组,每组18只,分别为假手术组(Sham组)、模型组(Model组)、静脉移植组(Vein组)、静脉移植+通心络组(Vein+TXL组)。小鼠MCAO模型采用改良线栓法制作;假手术组仅暴露、分离小鼠右侧颈部血管,不做其他操作;其余各组造模方法相同。小鼠脑缺血再灌注72 h通过尾静脉移植1×10^(5)个HUCMSCs,采用灌胃给药方式,静脉移植+通心络组小鼠每天的给药体积为10 mL·kg^(-1),模型组和假手术组给予等量的生理盐水。在移植后1、3、7天对各组小鼠进行取材,参照Longa评分法对小鼠进行神经功能评分,使用TTC染色法测定小鼠脑梗死面积。使用ELISA方法检测SDF-1α的表达,使用Werstern blot方法检测CXCR4的表达。最后采用SPSS 23.0统计软件对实验数据进行分析。结果(1)移植后的第1、3、7天,和Model组相比较,Vein组、Vein+TXL组小鼠神经行为学评分、梗死面积均降低,尤其是移植后第7天,与Model组比较,Vein组、Vein+TXL组神经功能评分、梗死面积差异有统计学意义(P<0.05)。(2)移植后第1、3、7天,与模型组相比较,静脉移植组、静脉移植+通心络组SDF-1α表达在移植后第1、3天升高,在第7天略降低,差异有统计学意义(P<0.05);移植后第1、3、7天,与Model组相比,Vein组、Vein+TXL组CXCR4表达在移植后第1、3天升高,在移植后第7天略降低,中药通心络干预后具有促进作用。结论静脉移植HUCMSCs可明显降低小鼠神经功能评分、减少脑梗死面积,上调SDF-1、CXCR4的表达,促进移植HUCMSCs向缺血组织迁移,修复神经功能缺损;静脉移植HUCMSCs可能通过SDF-1α/CXCR4生物轴参与改善脑缺血,中药通心络干预后具有促进作用。 Objective To explore the mechanism of intravenous transplantation of HUCMSCs through the SDF-1α/CXCR4 biological axis to improve cerebral ischemia,and to clarify the effect of the traditional Chinese medicine Tongxinluo capsule intervention.Methods The mice were randomly divided into 4 groups with 18 mice in each group,which were Sham group,Model group,Vein transplantation group,and Vein transplantation+Tongxinluo group.The mouse MCAO model was established by the modified suture method;In the sham operation group,only the right neck vessels of the mice were exposed and separated,and no other operations were performed;the modeling methods of the other groups were the same.At 72 hours after cerebral ischemia reperfusion,1×10^(5)hUCMSCs were transplanted through caudal vein and administered intragastric administration.The intravenous transplantation+Tongxinluo group was given10 mL·kg^(-1),once a day,and the model group and sham operation group were given the same amount of normal saline each time.Mice in each group were collected at 1,3 and 7 days after transplantation.The neurological function of mice was evaluated according to Longa score,and the cerebral infarct size of mice was measured by TTC staining.SDF-1αexpression was detected by ELISA and CXCR4 expression was detected by Western blot.Finally,SPSS 23.0 statistical software was used to analyze the experimental data.Results(1)Compared with the model group,the neurobehavioral scores and infarct area of the mice in the vein transplantation group,vein transplantation+Tongxinluo group were reduced on the 1,3,and 7 days after transplantation,especially on the 7thday after transplantation.Compared with the model group,the differences in nerve function scores and infarct area in the vein transplantation group,vein transplantation+Tongxinluo group were statistically significant(P<0.05).(2)Compared with the model group on day 1,3,and 7 after transplantation,the expression of SDF-1αin the vein transplantation group,vein transplantation+Tongxinluo group increased o
