基于信息熵赋权法的正交试验优化止血化痔合剂提取工艺 被引量:3

摘要 目的:优选止血化痔合剂的提取工艺。方法:采用HPLC法测定没食子酸、盐酸黄柏碱、黄芩苷的含量,采用紫外-可见分光光度法测定总黄酮的含量。在单因素试验的基础上,以溶剂倍量、浸泡时间、提取时间为考察因素,进行正交试验设计,以没食子酸、盐酸黄柏碱、黄芩苷和总黄酮的含量为评价指标,通过信息熵赋权法计算各指标的权重系数并进行加权处理得综合评分,采用综合评分进行正交分析优选最佳工艺。结果:止血化痔合剂的最佳提取工艺为14倍量水,浸泡1 h,提取2次,每次1 h。3批验证实验综合评分均值为98.31,RSD为1.97%(n=3)。结论:优选的止血化痔合剂的提取工艺重复性好、稳定可行,为该制剂的后续开发提供参考依据。 Objective:To optimize the extraction of Zhixue Huazhi mixture.Methods:An HPLC method was adopted to determine the contents of gallic acid,phellodendrine hydrochloride and baicalin.UV-Vis spectrophotometry was used to determine the content of total flavonoids.Based on single factor tests,orthogonal test was carried out to investigate the effects of solvent times,immersing time and extraction time on the extraction process.Using the contents of gallic acid,phellodendrine hydrochloride,baicalin and total flavonoids as the indexes,the weight coefficients of above indicator components were calculated by information entropy weighting,so as to calculate the comprehensive score.The optimal technology was obtained by orthogonal analysis based on the comprehensive score.Results:The optimal extraction technology of Zhixue Huazhi mixture was as follows:using 14-fold amount of water,immersing for 1 h,and extracting 2 h for twice.The mean comprehensive score of three batches of samples was 98.31 and the RSD was 1.97%.Conclusion:The optimal extraction technology of Zhixue Huazhi mixture is reproducible and stable,which provides reference for the subsequent development of the preparation.
