摘要
目的探讨DYS526a/b,DYS626,DYS713基因座在AGCU Y SUPP PLUS试剂盒与Goldeneye^(TM)Y ADD试剂盒片段长度多态性分型存在差异的原因。方法用上述两个试剂盒对9948标准品进行STR检测,并用9948标准品对上述4个基因座进行Sanger测序,根据文献报道、国际法医遗传学会Y-STR命名原则,以及国家公共安全行业标准对4个基因座的测序结果进行结构分析,按照不同的基序计算分型。结果9948标准品在上述两个试剂盒中的DYS526a/b,DYS626,DYS713基因座都存在分型差异。依据测序结果,根据DYS526a公认的基序(CCTT)_(a),分型应为13;根据行业标准《序列多态STR等位基因命名规则》中对DYS526b和DYS626确定的基序,分型均为25;DYS713的基序虽没有统一标准,但根据Y-STR命名指导意见,笔者认为9948的分型应为45。结论DYS526a/b,DYS626,DYS713基因座作为公安机关数据库建设备选基因座,在实际检案中经常被应用,试剂盒在研制过程中的严谨性和不同基序选择会使同一样品在不同试剂盒出现多个基因座分型差异,严重影响数据库比对效能。建议行业对产品加以管理,报批生产要提供基因座基序,统一不同产品相同基因座的基序,便于实战应用。
Objective To explore the reasons that there were differences in the nomenclature of DYS526a/b,DYS626and DYS713 by the AGCU Y SUPP PLUS kit and the Goldeneye^(TM)Y ADD kit.Methods The two kits were used to detect the typing results of 9948 standards.Sequenced the 9948 standards and analyzed the results according to literature reports,the Y-STR naming principles of the International Society for Forensic Genetics,and the national public safety industry standard.Then typed the four loci according to different motifs.Results The typing results for the DYS526a/b,DYS626,and DYS713 of the two kits were different.According to the sequencing results and the(CCTT)_(a)motif of DYS526a,its typing results should be 13.Referencing to the industry standard Sequence Polymorphic STR Allele Naming Rules,the typing results should be 25 for DYS526b and DYS626.Although there was no uniform standard for the motif of DYS713,according to the Y-STR naming guidelines,the typing results of the 9948 standards should be 45.Conclusion DYS526a/b,DYS626,and DYS713,as candidate loci for public security organ database construction,have been often used in forensic casework.The differences in the academic rigor and the selection of different motifs during kit design will make different typing results for the same sample,which will seriously affect the database comparison performance.It is recommendedthat the locus motifs should be provided for production approval and the motifs of the same locus should be unified,to facilitate their practical applications..
