摘要
目的探讨转化生长因子-β1(TGF-β1)、紧密连接蛋白(claudin-1)在胃息肉、胃癌(GC)中的表达水平及其与幽门螺杆菌(Hp)感染的关系。方法选取2019年1月至2021年5月本院收治的胃息肉患者65例为息肉组,GC患者62例为GC组,另选取行胃镜检查的健康者58例为对照组。测定胃黏膜组织Hp感染情况;实时荧光定量PCR(qRT-PCR)法测定胃黏膜组织TGF-β1、claudin-1 mRNA表达水平;Spearman法分析胃息肉、GC患者胃黏膜组织中TGF-β1 mRNA、claudin-1 mRNA表达水平与Hp感染的关系;Pearson法分析GC患者胃黏膜组织中TGF-β1 mRNA表达水平与claudin-1 mRNA的相关性。结果对照组、息肉组、GC组Hp阳性率、胃黏膜组织TGF-β1 mRNA表达水平呈升高趋势(P<0.05),claudin-1 mRNA表达水平呈降低趋势(P<0.05);息肉组、GC组Hp阳性的受试者TGF-β1 mRNA表达水平高于Hp阴性受试者(P<0.05),claudin-1 mRNA表达水平低于Hp阴性受试者(P<0.05);胃息肉、GC患者胃黏膜组织中TGF-β1 mRNA表达水平均与Hp感染呈正相关(P<0.05);胃息肉、GC患者胃黏膜组织中claudin-1 mRNA表达水平与Hp感染呈负相关(P<0.05);GC患者胃黏膜组织中TGF-β1 mRNA表达水平与claudin-1 mRNA呈负相关(P<0.05)。结论胃息肉、GC患者胃黏膜组织TGF-β1 mRNA呈高表达,claudin-1 mRNA呈低表达,两者均与Hp感染相关,检测Hp感染情况、测定胃黏膜组织TGF-β1 mRNA、claudin-1 mRNA表达水平有助于判断胃部病变程度,为临床防治胃息肉、GC提供新思路。
Objective To investigate expression levels of transforming growth factor-beta1(TGF-β1)and tight junction protein Claudin-1 in gastric polyps and gastric cancer(GC)and their relationships with Helicobacter pylori(Hp)infection.Methods From January,2019 to May,2021,65 patients with gastric polyps admitted to our hospital were selected as the polyp group,62 patients with GC were selected as the GC group,and 58 healthy people who underwent gastroscopy were selected as the control group.Hp infection in gastric mucosa tissue were measured;Real-time fluorescent quantitative PCR(qRT-PCR)method was applied to determine expression levels of TGF-β1 and claudin-1 mRNA in gastric mucosa tissue;Spearman method was conducted to analyze the relationship between expression levels of TGF-β1 mRNA and claudin-1 mRNA in gastric mucosal tissues of gastric polyps and GC patients and Hp infection;Pearson method was used to analyze the correlation between TGF-β1 mRNA expression level and claudin-1 mRNA in gastric mucosal tissues of GC patients.Results The positive rate of Hp and the expression level of TGF-β1 mRNA in gastric mucosal tissue of the control group,polyp group and GC group showed an increasing trend(P<0.05),while the expression level of claudin-1 mRNA showed a decreasing trend(P<0.05);TGF-β1 mRNA expression level of Hp-positive subjects in the polyp group and GC group was higher than that of Hp-negative subjects(P<0.05),while the expression level of claudin-1 mRNA was lower than that of Hp-negative subjects(P<0.05);The expression level of TGF-β1 mRNA in gastric mucosal tissues of gastric polyps and GC patients was positively correlated with Hp infection(P<0.05);The expression level of claudin-1 mRNA in gastric mucosal tissues of gastric polyps and GC patients was negatively correlated with Hp infection(P<0.05);The expression level of TGF-β1 mRNA in gastric mucosa of GC patients was negatively correlated with claudin-1 mRNA(P<0.05).Conclusion Gastric polyps and GC patients have a high expression of TGF-β1 mRNA and low
