摘要
目的了解克拉玛依市新甲型H1N1流感病毒HA基因特征。方法收集2018年—2020年新甲型H1N1流感毒株进行HA基因序列测定,运用Mega 6.06软件与疫苗株进行序列比对和分析。结果2018年—2019年毒株与疫苗株A/Michigan/45/2015核苷酸和氨基酸同源性分别为97.83%~98.62%和98.20%~99.10%。2020年毒株与疫苗株A/Brisbane/02/2018核苷酸和氨基酸同源性分别为98.53%~98.95%和98.22%~98.76%。有3株毒株在同一株内4个以上氨基酸位点较疫苗株发生替换,且涉及到2个以上抗原决定簇。所有毒株均有7个糖基化位点,裂解位点氨基酸序列均为V^(338) PSIQSR↓GLFGA,与疫苗株完全一致。结论流行毒株呈低致病性,2020年接种的流感疫苗较2018年—2019年接种的疫苗对克拉玛依市新甲型H1N1有更理想的预防效果。
Objective To understand the HA gene characteristics of new influenza A(H1N1)virus in Karamay City.Methods The HA gene sequences of new A/H1N1 influenza strains from 2018 to 2020 were determined,and the sequences were compared and analyzed with vaccine strains by Mega 6.06 software.Results The nucleotide and amino acid homologies in 2018—2019 strain and vaccine strain A/Michigan/45/2015 were 97.83%-98.62%and 98.20%-99.10%,respectively.The nucleotide and amino acid homologies of the 2020 strain and vaccine strain A/Brisbane/02/2018 were 98.53%-98.95%and 98.22%-98.76%,respectively.Three strains had more than four amino acid substitution sites in the same strain compared with the vaccine strain,and involved more than two epitope clusters.All strains had seven glycosylation sites,and the amino acid sequence of cleavage sites was V^(338) PSIQSR↓GLFGA,which was exactly the same as the vaccine strain.Conclusion The circulating strains were of low pathogenicity.Compared with the vaccine inoculated from 2018 to 2019,and the influenza vaccine inoculated in 2020 had a better preventive effect on new H1N1 in Karamay City.
