产β-甘露聚糖酶菌株筛选及制备甘露寡糖产物分析

摘要 从河南省济源市猪场仔猪粪便中筛选得到1株产β-甘露聚糖酶的菌株,通过外观形态学分析、生理生化结果分析、16S rDNA序列分析及构建系统发育树结果,鉴定该菌株为沙福芽孢杆菌(Bacillus safensis),命名为WBT0011。通过β-甘露聚糖酶和魔芋胶、瓜尔豆胶制备甘露寡糖,发现魔芋胶和瓜尔豆胶降解为多种甘露寡糖产物,说明二者水解较为完全,本研究旨在为β-甘露聚糖酶的应用及进一步工业化生产甘露寡糖的工艺优化提供帮助。 A strain producingβ-mannanase was screened from the feces of piglets in Jiyuan city,Henan province.The strain was identified as Bacillus safensis and named WBT0011 by morphological analysis,physiological and biochemical analysis,16S rDNA sequence analysis and phylogenetic tree construction.Mannan oligosaccharides were prepared byβ-mannanase,konjac glucomannan and guar gum.It was found that konjac glucomannan and guar gum were degraded into a variety of oligosaccharide products,indicating that they were hydrolyzed completely.It is hoped to provide help for the application ofβ-mannanase and the optimization of industrial production of mannan oligosaccharides.
