摘要
目的:探讨银屑平丸联合阿维A胶囊对银屑病样小鼠模型的疗效及血清炎性因子的影响。方法:将40只BALB/c小鼠(SPF级,雌雄各半)按照完全随机原则分为空白对照组、模型组、阿维A胶囊组、银屑平丸组、阿维A胶囊+银屑平丸组(中西结合组)5组,每组8只。空白对照组小鼠每日用白凡士林涂抹背部暴露皮肤,其他各组小鼠则用5%咪喹莫特乳膏涂抹背部暴露皮肤,每日1次,连续涂抹8日。抹药同时给各组小鼠通过灌胃针进行灌胃干预,连续用药8日。实验过程中每天对每组小鼠背部皮损变化进行拍照存档。末次用药24h后,观察每组小鼠皮损变化并予以皮损面积及严重程度指数(PASI)评分,采用酶联免疫吸附测定(ELISA)法检测各组小鼠血清IL-17、IL-23的表达水平,后续剪取银屑病小鼠皮损组织予切片、HE染色以观察病理组织变化。结果:与空白对照组比较,模型组、阿维A胶囊组、银屑平丸组、中西结合组小鼠背部皮损的PASI评分均显著升高(P<0.05);与模型对照组相比,阿维A胶囊组、银屑平丸组、中西结合组小鼠背部皮损的PASI评分均显著降低(P<0.05),其中中西结合组疗效最佳(P<0.05);HE染色结果亦证实中西结合组的缓解程度最佳(P<0.05);ELISA结果显示,阿维A胶囊组、银屑平丸组、中西结合组小鼠血清IL-17、IL-23水平均显著降低,其中中西结合组效果最佳(P<0.05)。结论:银屑平丸联合阿维A胶囊可明显改善PS小鼠模型的皮损状况,均较单独使用银屑平丸或阿维A胶囊疗效更佳,其作用机制可能与抑制PS小鼠模型血清IL-17、IL-23的表达水平相关。
Objective:To investigate the curative effect of Yinxieping Pill combined with Acitretin capsules on psoriasislike mouse model and the effect of serum inflammatory factors.Methods:Forty BALB/c mice(SPF grade,half male and female)were divided into blank control group,model control group,Avitamin A capsule group,Yinxieping pill group,Avitamin A capsule and Yinxieping pill according to the principle of complete randomization.Group(combined medication group)and other 5 groups,each with 8 animals.The mice in the blank control group were smeared with white petrolatum on the back exposed skin every day,and the mice in the other groups were smeared with 5%imiquimod cream on the back exposed skin,once a day for 8 consecutive days.The smear was given to each group of mice at the same time through intragastric gavage needle for intragastric intervention for 8 consecutive days.During the experiment,the back skin lesions of each group of mice were photographed and archived every day.24 hours after the last medication,observe the changes in the photos of the skin lesions of each group of mice and score them with PS area and severity index(PASI),and detect by enzyme linked immunosorbent assay(ELISA)method The expression levels of serum IL-23,IL-17 and other inflammatory factors in the mice of each group were subsequently cut out and sliced and HE stained to observe the pathological changes in the skin lesions of the psoriasis mice.Results:Compared with the blank control group,the severity of back skin lesions in the model control group,the Acitretin capsule group,the Yinxieping pill group,and the combination medication group was significantly increased by calculating the PASI score(P<0.05);and the model Compared with the control group,the severity of back skin lesions in the Acitretin capsule group,Yinxieping pill group,and combination medication group was significantly reduced by calculating the PASI score(P<0.05),and the combination medication group had the best effect(P<0.05);HE staining results also confirmed that the combin
