消肿喷剂主药对体外炎症模型小鼠白细胞介素及NLRP3相关蛋白表达影响的研究

摘要 目的:研究消肿喷剂促进急性骨骼肌损伤炎症修复的作用机制,及炎症在急性骨骼肌损伤修复过程中的作用机制。方法:采用脂多糖(LPS)诱导C2C12小鼠成肌细胞,建立体外细胞炎性模型,ELISA试剂盒检测C2C12细胞上清中白细胞介素(IL-1β、IL-18)含量,qRT-PCR检测C2C12细胞ASC、Caspase-1、NLRP3基因表达,Western blot检测ASC、Caspase-1、NLRP3蛋白的表达情况。结果:NO抑制剂(L-NAME)、NO供体(SNP)能够明显提高或降低IL-1β、IL-18,ASC、Caspase-1和NLRP3 mRNA表达水平和蛋白水平,消肿喷剂主药含药血清能够显著降低IL-1β、IL-18及ASC、Caspase-1、NLRP3 mRNA表达水平和蛋白水平。结论:LPS激活NO-NLRP3-IL炎症轴,诱导建立C2C12细胞体外炎症模型。NO通过NO/ASC/Caspase-1/NLRP3/IL-1β/IL-18轴,调控骨骼肌体外炎症模型炎症因子表达,消肿喷剂可能通过干预NO表达,减轻炎性反应,促进骨骼肌损伤修复。 Objective:To study the mechanism of detumescence spray promoting the inflammatory repair of acute skeletal muscle injury and the mechanism of inflammation in the process of acute skeletal muscle injury repair.Methods:LPS induced C2C12 mouse myoblasts were used to create an in vitro inflammatory model.IL-1βand IL-18 levels in superserum of C2C12 cells were detected by ELISA kit.The expression of ASC,Caspase-1 and NLRP3 genes in C2C12 cells was detected by qRT-PCR and the expression of ASC,Caspase-1 and NLRP3 proteins was detected by Western blot.Results:L-NAME and SNP can significantly increase or decrease IL-1β,IL-18,ASC,Caspase-1 and NLRP3 mRNA expression levels and protein levels.The main drug containing serum of detumescence spray can significantly reduce the mRNA expression levels of IL-1β,IL-18,ASC,Caspase-1,NLRP3 and protein levels.Conclusion:LPS-induced inflammatory model of C2C12 cells in vitro activates the NO-NLRP3-IL inflammatory axis,and NO regulates the expression of inflammatory factors in skeletal muscle inflammatory model through the NO/ASC/Caspase-1/NLRP3/IL-1β/IL-18 axis.Detumescence spray may reduce inflammation and promote skeletal muscle injury repair by interfering with NO expression.
