摘要
目的采用组合筛选思路探讨黄连解毒汤中小檗碱、黄芩苷和栀子苷不同模拟组合对高血脂症模型小鼠血脂代谢的影响。方法将C57BL/6J小鼠以高脂饮食诱导建立高血脂症模型,实验分为正常组、模型组、阳性药物组、黄连解毒汤组、黄连解毒汤模拟药物组、模拟药物小檗碱高剂量组、模拟药物黄芩苷高剂量组和缺栀子苷模拟药物组,分别连续灌胃给药4周后取血,检测TC、TG、LDL-C、Lp-PLA2、SOD、LCAT等指标。结果与模型组比较,黄连解毒汤及其模拟药物均可降低血脂指标。增加模拟药物中小檗碱含量能明显降低TC、TG、LDL以及Lp-PLA2等指标,并明显提高SOD、LCAT的表达(P<0.05)。模拟药物黄芩苷高剂量组和缺栀子苷模拟药物组TC、TG、Lp-PLA2均升高,同时SOD、LCAT的表达降低(P<0.05)。结论小檗碱是调节血脂代谢的关键成分,其作用可能与提高SOD、LCAT的表达相关。增加黄芩苷的含量则不利于调节血脂代谢,栀子苷的调节作用则可促进复方给药作用的最大化,三者间的配比优化有待进一步研究。
Objective To investigate the effects of different simulated combinations of berberine,baicalin and geniposide in Huanglian Jiedu decoction on blood lipid metabolism in hyperlipidemia model mice by combination screening.Methods C57BL/6J mice are induced by high-fat diet to establish hyperlipidemia model.The mice are divided into normal control group,model group,positive drug group,Huanglian Jiedu decoction group,Huanglian Jiedu decoction simulated drug group,berberine high dose group,baicalin high dose group and simulated drug group lacking geniposide.After 4 weeks of continuous intragastric administration,the blood samples are taken to detect the levels of TC,TG,LDL-C,Lp-PLA2,SOD and LCAT.Results Compared with the model group,Huanglian Jiedu decoction and its simulated drugs could reduce blood lipid indexes,while increasing the content of berberine could significantly reduce TC,TG,LDL and Lp-PLA2,and significantly increase the expression of SOD and LCAT(P<0.05).In the high dose group of baicalin and simulated drug group lacking geniposide,TC,TG and Lp-PLA2 increased significantly,while the expressions of SOD and LCAT decreased significantly(P<0.05).Conclusion The increase of berberine in the simulated drugs of Huanglian Jiedu decoction can significantly reduce the blood lipid index,which is the key component of regulating the metabolism of blood lipid,and its effect may be related to the improvement of SOD and LCAT expression.Increasing the content of baicalin is not conducive to regulating blood lipid metabolism,and the regulatory effect of geniposide can promote the maximization of compound administration.The optimization of the proportion of the three needs to be further studied.
