黄粉虫多肽的组成及体外抗氧化活性 被引量:4

Composition and In-Vitro Antioxidant Activity of Polypeptide from Tenebrio Molitor

摘要 以碱性蛋白酶酶解后的黄粉虫多肽为原料,利用透析袋将其按相对分子质量(Mr)分为小于1000 u、1000~2000 u、2000~3500 u和大于3500 u 4个组分,考察了黄粉虫多肽的氨基酸组成和分子质量分布,并对各组分体外抗氧化活性进行研究。结果表明,黄粉虫多肽中与抗氧化相关的氨基酸含量较高(疏水性氨基酸和带电氨基酸分别为33.31%和31.14%),小于1000 u的组分含量(63.71%)显著高于其他组分(P<0.05);多肽的体外抗氧化能力与分子质量大小密切相关,其中大于3500 u组分的DPPH自由基清除能力最强(IC_(50)值为11.61 mg/mL),1000~2000 u组分的总还原力(5 mg/mL时A700为1.13)及.O_(2)^(-)自由基清除能力最强(IC_(50)值为2.36 mg/mL)。综上黄粉虫多肽中1000~2000 u的组分抗氧化活性最强,可为抗氧化肽后续分离纯化及抗氧化功能食品的开发提供数据支撑。 Tenebrio Molitor polypeptide obtained by alkaline protease was used as a raw material.It was separated according to relative molecular mass into four components by the dialysis bag,which is less than 1000 u,1000~2000 u,2000~3500 u and greater than 3500 u.The amino acid composition and relative molecular weight distribution of the Tenebrio Molitor polypeptide were measured,and the in vitro antioxidant activity of each component was studied.The results showed that the amino acid content associated with antioxidant activity in Tenebrio Molitor polypeptide was high(The hydrophobic amino acid and the charge amino acid content were 33.31% and 31.14%,respectively),and the component content of less than 1000 u(63.71%) significantly higher than other components(P<0.05).The in vitro antioxidant capacity was closely related to the molecular weight,wherein the DPPH scavenging activity of greater than 3500 u components was the strongest(IC_(50) was 11.61 mg/mL).The total reducing power(5 mg/mL,A700 1.13)and the.O_(2)^(-) scavenging activity(IC_(50) was 2.36 mg/mL) of 1000~2000 u components was the strongest.In summary,component of 1000~2000 u in Tenebrio Molitor polypeptides had the strongest antioxidant activity,and the data support could be provided for subsequent separation purification of antioxidant peptides the development of antioxidant functional food.
