鱼腥草对金黄色葡萄球菌生物被膜干预作用研究 被引量:4

摘要 研究鱼腥草水提物对金黄色葡萄球菌生物被膜的干预作用。采用微量肉汤稀释法测定鱼腥草水提物对金黄色葡萄球菌CMCC(B)26003的最低抑菌浓度;采用结晶紫染色法、扫描电镜及激光共聚焦显微镜评估鱼腥草水提物对CMCC(B)26003生物被膜形成的影响;最后采用实时荧光定量PCR方法测定鱼腥草水提物对O-乙酰丝氨酸硫化氢裂解酶B(O-acetylserine sulfhydrylase-B)cysM基因转录的影响。结果显示,鱼腥草水提物在12.500 mg/mL、6.250 mg/mL、3.125 mg/mL质量浓度下均可以有效抑制金黄色葡萄球菌生物被膜的形成,在12.500 mg/mL质量浓度下可下调cysM基因的表达。鱼腥草可能通过下调cysM基因的表达而抑制金黄色葡萄球菌生物被膜的形成。 In order to provide the basis for developing anti-biofilm drugs and controlling the cow mastitis and other diseases caused by Staphylococcus aureus biofilm infection,the effect of aqueous extract from Herba Houttuyniae on Staphylococcus aureus biofilm formation was studied.The minimal inhibitory concentration(MIC)of aqueous extract from Herba Houttuyniae against Staphylococcus aureus CMCC(B)26003 was determined by broth microdilution method.The inhibitory effect of the aqueous extract on the CMCC(B)26003 biofilm formation was evaluated by crystal violet staining,scanning electron microscopy and confocal laser scanning microscopy.Finally,the effect of Herba Houttuyniae on the transcription of cysM was determined by real-time quantitative PCR.The results showed that aqueous extract from Herba Houttuyniae could inhibit the Staphylococcus aureus biofilm formation effectively at the concentration of 12.500 mg·mL^(-1),6.250 mg·mL^(-1)and 3.125 mg·mL^(-1),and down-regulate the expression of cysM gene at the concentration of 12.500 mg·mL^(-1).Herba Houttuyniae may inhibit the Staphylococcus aureus biofilm formation by down-regulating the expression of cysM gene.
