摘要
目的探讨胰岛素样生长因子结合蛋白4(IGFBP-4)在非小细胞肺癌中的表达及作用。方法选取丽水市中心医院2008年1月至2020年1月行手术治疗的20例非小细胞肺癌患者的肺癌组织以及正常肺组织,检测上述标本中IGFBP-4的表达情况。将非小细胞肺癌A549细胞分为Control组和不同浓度IGFBP-4(5、50、500 ng/ml)处理组。采用CCK-8法绘制细胞生长曲线,平板克隆实验计算细胞集落数量,观察IGFBP-4对细胞增殖的影响,Transwell实验检测IGFBP-4对细胞迁移和侵袭能力的影响,Western blot法检测IGFBP-4处理后细胞内Fibronectin和c-fos蛋白表达水平。结果IGFBP-4在肺癌组织中的表达明显下调。经IGFBP-4处理的A549细胞的增殖活性、迁移和侵袭能力均明显低于Control组(均P<0.05),Fibronectin和c-fos蛋白表达水平也明显下调(均P<0.05)。结论IGFBP-4在非小细胞肺癌组织中的表达明显下调,可抑制A549细胞的增殖、迁移和侵袭,并降低Fibronectin和c-fos蛋白的表达水平,表现出较强的抗肿瘤作用。
Objective To investigate the effect of insulin-like growth factor binding protein-4(IGFBP-4)on proliferation and migrant ability of non-small cell lung cancer(NSCLC)cells.Methods The expression of IGFBP-4 was detected in 20 samples of NSCLC and normal lung tissue.NSCLC A549 cells were treated with different concentrations(5,50,500 ng/ml)of IGFBP-4.The cell proliferation was detected by CCK-8 method and the clone formation rate was measured by plate cloning assay,cell migration and invasion ability was determined by Transwell assay,the expression levels of Fibronectin and c-fos proteins were tested by Western blot.Results Compared with normal lung tissue,IGFBP-4 expression in lung cancer tissues was downregulated.The proliferative activity,migration and invasion ability of IGFBP-4 treated A549 cells were significantly lower than those of the control cells(P<0.05).IGFBP-4 reduced Fibronectin and c-fos expression.Conclusion IGFBP-4 expression is significantly downregulated in non-small cell lung cancer.IGFBP-4 can inhibit the proliferation,migration and invasion of NSCLC A549 cells and decrease expression of Fibronectin and c-fos proteins,which indicates that IGFBP-4 has potent antitumor effects in non-small cell lung cancer.
