中蜂蜂毒和意蜂蜂毒抑菌作用分析 被引量:3

摘要 为开发无耐药性的传统抗生素的补充抗菌剂,试验以中蜂和意蜂蜂毒为研究对象,通过琼脂平板扩散法和最小抑菌浓度法(MIC)检测二者对大肠杆菌、金黄色葡萄球菌、白色葡萄球菌、枯草芽孢杆菌、苏云金芽孢杆菌、伤寒杆菌、无乳链球菌、沙门氏菌和尖孢镰刀菌等8种病原细菌和1种真菌的抑制作用,同时比较2种蜂毒的抑菌活性差异;采用酶标仪和荧光显微镜试验验证蜂毒对无乳链球菌的抑制功效。结果表明,中蜂蜂毒、意蜂蜂毒对供试革兰氏阳性菌、真菌和部分革兰氏阴性菌都有抑制作用,随着蜂毒质量浓度的升高,抑菌圈直径逐渐增大,抑菌效果愈加明显;中蜂蜂毒和意蜂蜂毒对革兰氏阳性菌和真菌的最小抑菌质量浓度(MIC)均为5 mg/mL。酶标仪检测数据显示,阴性对照组在培养无乳链球菌12 h后OD值达到0.531,而质量浓度为0.5、2.5、5.0 mg/mL的意蜂蜂毒OD值仅为0.153、0.185、0.196。通过正置荧光显微镜观察发现,空白对照组有大量细菌繁殖,而青霉素组和蜂毒组中无细菌存在。研究进一步丰富和明确了蜂毒的抑菌特性,且证实了中蜂蜂毒和意蜂蜂毒二者的抑菌效果相当。 To provide basic data for the development of a traditional antibiotics with non-drug-resistance as a supplement of antimicrobial agent,in this study,bee venom(BV)of of A.cerana cerana and A.mellifera ligustica,were selected as objects studied,by AGAR plate diffusion method and minimum inhibitory concentration(MIC),bacteriostasis of 8 pathogenic bacteria and 1 fungus including Escherichia coli,Staphylococcus aureus,Staphylococcus albus,Bacillus subtilis,Bacillus thuringiensis,Salmonella typhi,Streptococcus agalactiae,Salmonella and Fusarium oxysporum was detected and the differences of bacteriostatic activity between the two kinds of BV were compared.A microplate reader and a fluorescence microscope were used to verify the inhibitory effect of BV on Streptococcus agalactiae.The results showed the BV of A.cerana cerana and A.mellifera ligustica had inhibitory effect on Gram-positive bacteria,fungi and some Gram-negative bacteria.As the concentration of BV increased,the diameter of the bacteriostatic zone increased,and the bacteriostatic effect became more obvious.The minimum inhibitory concentrations(MIC)of BV of A.cerana cerana and A.mellifera ligustica against Gram-positive bacteria and fungi were both 5 mg/mL.The detecting data of the microplate reader showed that the OD value of the negative control group reached0.531 after 12 h of culture,while the OD values of the BV with concentrations of 0.5,2.5,5.0 mg/mL were only 0.153,0.185,0.196.Observation through an upright fluorescence microscope revealed that a large number of bacteria reproduced in the blank control group,but no bacteria existed in the penicillin group and the BV group.This study further enriched and clarified the antibacterial properties of BV,and confirmed that the bee venom had bacteriostatic effect,and the BV from A.cerana cerana and A.mellifera ligustica had the same bacteriostatic effect.
