响应面法优化ε-聚赖氨酸产生菌的发酵培养基 被引量:3

Optimization of Fermentation Medium ofε-Polylysine-Producing Bacteria by Response Surface Methodology

摘要 为了提高ε-聚赖氨酸的产量,对ε-聚赖氨酸产生菌突变菌株进行了更经济、产量提高效果更好的碳、氮源筛选并应用响应面试验优化了发酵培养基配方。对筛选出的碳源、氮源分别进行复配,确定比例后,通过响应面试验优化结果为:复合碳源总浓度为40 g/L,碳源配比为葡萄糖∶糖蜜为1∶4,复合氮源浓度为15 g/L,氮源配比为氯化铵∶玉米浆为1∶2。最终发酵培养基配方为:葡萄糖8 g/L,糖蜜32 g/L,氯化铵5 g/L,玉米浆10 g/L,MgSO_(4)·7H_(2) O 3.2 g/L,FeSO_(4)·7H_(2) O 0.08 g/L,ZnSO_(4)·7H_(2)O 0.04 g/L,K_(2)HPO_(4)2.4 g/L,KH_(2) PO_(4)3.2 g/L,此发酵培养基条件下产物产量维持在14.76 g/L左右。结果表明该试验可以降低碳源使用量,并用低成本的糖蜜代替了一部分的葡萄糖,以达到更经济并能够提高产量的效果。 In order to increase the yield ofε-polylysine,theε-polylysine-producing mutant strains are screened for carbon and nitrogen sources that are more economical and efficient,and the fermentation medium formula is optimized by response surface test.The selected carbon and nitrogen sources are compounded separately.After determining the proportions,the optimization results through the response surface test are as follows:the total concentration of the compound carbon source is 40 g/L,the carbon source ratio is glucose∶molasses of 1∶4,and the compound nitrogen source concentration is 15 g/L,and the nitrogen source ratio is ammonium chloride∶corn steep liquor of 1∶2.The final fermentation medium formula is glucose 8 g/L,molasses 32 g/L,ammonium chloride 5 g/L,corn steep liquor 10 g/L,MgSO_(4)·7H_(2) O 3.2 g/L,FeSO_(4)·7H_(2) O 0.08 g/L,ZnSO_(4)·7H_(2)O 0.04 g/L,K_(2)HPO_(4)2.4 g/L,KH_(2) PO_(4)3.2 g/L,the product yield is maintained at about 14.76 g/L under such fermentation medium conditions.The experimental results show that this experiment can reduce the usage amount of carbon source,and replace a part of glucose with low-cost molasses,in order to achieve a more economical effect and increase the yield.
