摘要
目的:探讨性别差异对小鼠溃疡性结肠炎(UC)模型成模的影响,为实验造模提供参考依据。方法:将24只C57BL/6J小鼠根据性别随机分为4组,雌性对照组、雄性对照组、雌性DSS组和雄性DSS组,每组6只,对照组正常饮食,雌性或雄性DSS组给予正常饮食+3%葡聚糖硫酸钠(DSS)溶液自由饮用7 d进行造模。每日记录各组小鼠精神状态、毛色光泽、体质量、进食量、粪便性状及隐血,评估疾病活动指数(DAI)、病死率、成模率、结肠长度、脾脏系数,苏木精-伊红(HE)染色观察结肠病理变化。结果:雌性DSS组小鼠DAI评分、病死率均较雄性DSS组低,脾脏系数较雄性DSS组低(P<0.01),体质量变化、结肠长度缩短较雄性DSS组较少(P<0.01,P<0.05),成模率较雄性DSS组高,结肠病理变化较雄性DSS组黏膜充血水肿轻,炎症细胞较少。结论:3% DSS溶液诱导C57BL/6J小鼠UC模型存在一定的性别偏倚,提示使用3% DSS溶液7 d建立雌性UC小鼠更易于模型的诱导。
Objective:To explore the influence of gender difference on UC modeling and provide reference for experimental modeling.Methods:Twenty-four C57BL/6J mice were randomly divided into four groups according to sex:female control group,male control group,female DSS group and male DSS group,with 6 mice in each group.The control group was given normal diet,and the female or male DSS groups were given normal diet +3% DSS for modeling for 7 days.Recorded body weight,mental state,hair color and luster,food intake,fecal traits and occult blood of mice in each group everyday,and evaluated DAI score,mortality rate,model rate,colon length,spleen coefficient;pathological changes of colon were observed by hematoxylin-eosinstaining.Results:Compared with the male DSS group,the DAI score and mortality of female DSS group were lower,spleen coefficient of female DSS group was lower (P<0.01),body weight change and colon length shortening were less (P<0.01,P<0.05),model formation rate was higher.Compared with the male DSS group,the pathological changes of colon were lighter,mucosal hyperemia and edema,and inflammatory cells were less.Conclusion:There is a gender bias in the UC model of C57BL/6J mice induced by 3% DSS solution,suggesting that it is easier to induce the female UC model of C57BL/6J mice induced by 3% DSS solution for 7 days.
