摘要
目的 探讨磷酸二酯酶3A(Phosphodiesterase3A,PDE3A)与子痫前期发病的相关性,为进一步完善子痫前期发病的分子机制研究提供数据支持。方法 选取2021年1月至10月在广州医科大学附属第三医院产科收治的30例子痫前期孕妇作为实验组,同期收治的30例健康妊娠孕妇作为对照组。首先通过苏木精-伊红(hematoxylin-eosin staining,H-E)染色观察胎盘组织内部的病理变化差异;然后采用免疫印迹(Westernblot,WB)和免疫组化(immunohistochemistry,IHC)实验分析PDE3A蛋白在胎盘组织中的表达差异,最后结合定量聚合酶链反应(quantitative polymerase chain reaction,qPCR)法检测子痫前期患者胎盘组织PDE3A mRNA表达水平。结果 H-E染色结果显示正常孕妇胎盘组织内部细胞清晰并排列整齐,绒毛组织几乎无破损。子痫前期孕妇的胎盘组织内部出现细胞溶解,血管水肿破裂。Western blot和IHC实验结果显示,与对照组相比,实验组中PDE3A蛋白表达显著增加,差异有统计学意义(P<0.05)。qPCR检测结果发现实验组中PDE3A mRNA表达量(1.48±0.23)大于对照组(0.12±0.04),差异有统计学意义(P<0.05)。结论 磷酸二酯酶3A在子痫前期患者胎盘组织中的表达水平高于正常孕妇胎盘组织,说明磷酸二酯酶3A可能参与子痫前期疾病的发生发展。
Objective To explore the correlation between phosphodiesterase 3A(PDE3A) and the pathogenesis of preeclampsia,and to provide data support for further improving the molecular mechanism of the pathogenesis of preeclampsia.Methods Thirty cases of preeclampsia pregnant women admitted to department of obstetrics,the Third Affiliated Hospital of Guangzhou Medical University from January to October 2021 were selected as the experimental group,and 30 cases of healthy pregnant women admitted during the same period were selected as the control group.First,the pathological changes in placental tissue were observed by H-E staining.Then Western blot and IHC were used to analyze the expression differences of PDE3A protein in placental tissues.Finally,qPCR was used to detect the expression levels of PDE3A mRNA in placental tissues of preeclampsia patients.Results The results of H-E staining showed that the cells in the placental tissue of normal pregnant women were clear and neatly arranged,and the villi tissue was almost undamaged.In women with preeclampsia,the placental tissue develops cytolysis and blood vessels rupture with edema.Western blot and IHC results showed that PDE3A protein expression was significantly increased in the experimental group compared with the control group,and the difference was statistically significant(P<0.05).qPCR results showed that PDE3A mRNA expression in the experimental group(1.48±0.23) was higher than that in the control group(0.12±0.04),and the difference was statistically significant(P<0.05).Conclusion The expression level of phosphodiesterase 3A in placental tissues of preeclampsia patients was higher than that of pregnant women,suggesting that phosphodiesterase 3A may be involved in the pathogenesis of preeclampsia.
