摘要
目的观察"皮粘散"对大鼠慢性皮肤创面组织中胶原蛋白(collagen)Ⅰ、Ⅳ表达的影响。方法 SPF级SD大鼠24只,随机分为模型组、象皮组、皮粘散组、硼砂组(外用药物0.2 g)。通过冰醋酸制造慢性皮肤溃疡大鼠模型,观察皮粘散对促进大鼠慢性皮肤创面愈合的作用,局部大鼠创面组织使用免疫荧光法(IF)检测胶原蛋白Ⅰ、Ⅳ的表达,计算溃疡面积积分。结果与模型组、象皮组、硼砂组相比,皮粘散组阳性荧光信号强,荧光染色所见胶原蛋白Ⅰ排列一致性较好;皮粘散组胶原蛋白Ⅳ阳性荧光信号强,荧光染色显示大量管腔类结构,可见完整连续的表皮组织基底膜结构;同时对大鼠慢性皮肤创面有良好的愈合作用。结论皮粘散对大鼠慢性创面有较好的治疗作用,可减少分泌物,促进创面愈合,其作用机制与上调大鼠创面组织中胶原蛋白Ⅰ、Ⅳ的表达有关。
Objective To observe the effect of “Pi Zhan San” on the expression of collagen Ⅰ and Ⅳ in chronic skin ulcers in rats.Methods Twenty-four SPF SD rats were randomly divided into model group, elephant skin group, Pi Zhan San group, borax group(external drug 0.2 g).A rat model of chronic skin ulcer was made by glacial acetic acid to observe the effect of Pi Zhan San on promoting the healing of chronic skin ulcers in rats.Immunofluorescence(IF) was used to detect the expression of collagen Ⅰ and Ⅳ in local rat ulcer tissues to calculate the ulcer area, points for color, secretion, skin lesions and odor.Results Compared with the model group, elephant skin group, and borax group, the Pi Zhan San group had a stronger positive fluorescence signal, and the collagen Ⅰ arrangement was better in the fluorescence staining;the Pi Zhan San group had a stronger collagen Ⅳ positive fluorescence signal, which was indicated by fluorescence staining.A large number of luminal structures, showing a complete and continuous basement membrane structure of epidermal tissue;at the same time, it had a good healing effect on chronic skin ulcers in rats.Conclusion Pi Zhan San has a good therapeutic effect on chronic skin ulcers.It can reduce secretions and promote ulcer healing.Its mechanism of action is related to the up-regulation of collagen Ⅰ and Ⅳ expression in rat ulcer tissues.
