基于网络药理学对附子治疗非酒精性脂肪性肝病作用靶点的研究

摘要 目的:运用网络药理学的方法对附子治疗非酒精性脂肪性肝病的主要活性成分及靶点进行研究。方法:通过中药药理数据库和分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)获取附子的化学成分,在Pub Chem下载其2D结构式,导入Pharm Mapper获取靶点蛋白,使用Unipro数据库将其转换为基因名。通过Gene Cards、人类孟德尔遗传综合数据库(OMIM)和TTD收集非酒精性脂肪性肝病相关疾病基因,对药物作用基因及疾病基因进行韦恩分析,寻找交集靶点,用交集靶点与对应活性成分构建活性成分-靶点网络,并用Divad平台进行富集分析。结果:得到作用于非酒精性脂肪性肝病的15个活性成分和73个交集靶点,以错误发现率<0.05为筛选条件,基因本体(GO)功能富集分析确定了57个条目,京都基因和基因组数据库(KEGG)通路富集分析发现17条作用通路。结论:本研究初步探讨了附子治疗非酒精性脂肪性肝病的作用机制,并为进一步的试验研究奠定良好基础。 Objective:To study the main active ingredients and targets of aconite in the treatment of non-alcoholic fatty liver disease by means of network pharmacology.Methods:The chemical composition of aconite were obtained from TCMSP database,its 2D structural formula was downloaded in PubChem and imported into PharmMapper to obtain target protein,and Unipro database was used to convert it into gene name.NAFLD-related disease genes were collected through GeneCards database,OMIM database and TTD database,a Vnne analysis on drug action genes and disease genes was performed to find intersection targets,intersection targets and corresponding active ingredients was used to construct an active ingredient-target network.And the Divad platform was used for enrichment analysis.Results:15 active ingredients and 73 overlapping targets that act on NAFLD were obtained.With FDR<0.05 as the screening condition,the GO function enrichment analysis identified 57 items,and the KEGG pathway analysis found 17 pathways.Conclusion:The results of this study preliminarily discussed the mechanism of action of aconite in treating NAFLD,and laid a good foundation for further research.
