摘要
目的:基于Wnt信号通路探讨血府逐瘀汤对非小细胞肺癌细胞生长的抑制作用及对β-连环蛋白(β-catenin)、p-糖原合成酶激酶3β(p-GSK-3β)蛋白表达情况的影响。方法:体外培养人肺癌细胞PC9细胞株,选取对数生长期的人肺癌PC9细胞,分为空白对照组、血府逐瘀汤15 mg/mL、30 mg/mL、45 mg/mL组。CCK-8法检测血府逐瘀汤对细胞生长的抑制作用,免疫荧光法检测各组细胞内p-GSK-3β与β-catenin蛋白的表达情况。结果:不同处理时间(24 h、48 h、72 h)的血府逐瘀汤15 mg/mL、30 mg/mL、45 mg/mL组抑制率均高于空白对照组(P <0.05),且随着时间与浓度的增加抑制效果更为显著。不同浓度血府逐瘀汤组β-catenin、p-GSK-3β蛋白表达水平均低于空白对照组(P <0.05),且随着浓度的上升下调水平更大。结论:血府逐瘀汤对人肺癌PC9细胞的增殖具有抑制作用,且呈现出浓度与时间依赖性;其抑制细胞增殖的机制可能与下调p-GSK-3β、β-catenin蛋白表达,阻断Wnt信号通路有关。
Objective:Based on the Wnt signaling pathway,the inhibitory effect of the Xuefu Zhuyu decoction(血府逐瘀汤)on the growth of non-small cell lung cancer cells,and its influences onβ-catenin and p-GSK-3βprotein expression levels were explored.Methods:Human lung cancer PC9 cell line was cultured in vitro,and human lung cancer PC9 cells in logarithmic growth phase were selected and divided into the blank control group,and the 15 mg/mL,30 mg/mL,45 mg/mL groups of the Xuefu Zhuyu decoction.The CCK-8 method was used to detect the inhibitory effect of the Xuefu Zhuyu decoction on cell growth,and the immunofluorescence method was used to detect the expression of p-GSK-3βandβ-catenin protein in the cells in each group.Results:The inhibition rates at different treatment times(24 h,48 h,72 h)in the 15 mg/mL,30 mg/mL,and 45 mg/mL groups of the Xuefu Zhuyu decoction were higher than those in the blank control group(P<0.05),and with the increase of time and concentration,the inhibitory effect was more significant.The protein expression levels ofβ-catenin and p-GSK-3βin groups of the Xuefu Zhuyu decoction at different concentrations were all lower than the blank control group(P<0.05),and the degrees of down-regulation were greater as the concentration increased.Conclusion:The Xuefu Zhuyu decoction has an inhibitory effect on the proliferation of human lung cancer PC9 cells,and shows a concentration and time dependence.Its mechanism of inhibiting cell proliferation may be related to down-regulation of p-GSK-3βandβ-catenin protein expression and blocking Wnt signaling access.
