不同浓度薏苡仁提取物对口腔鳞状细胞癌细胞增殖、迁移和侵袭的影响及作用机制 被引量:7

Effects of Different Concentrations of Coix Seed Extract on Proliferation,Migration and Invasion of Oral Squamous Cell Carcinoma Cells and Its Mechanism

摘要 目的探讨不同浓度薏苡仁提取物对口腔鳞状细胞癌(OSCC)细胞增殖、迁移和侵袭的影响及作用机制。方法体外培养人OSCC CAL27细胞,按照实验设计分为高剂量组、低剂量组、对照组,高、低剂量组分别予20、10μmol/L薏苡仁提取物干预,对照组不予任何干预。各组细胞培养24 h后采用MTT法、细胞划痕法、Transwell法分别检测细胞增殖率、迁移能力、体外侵袭能力,同时采用蛋白免疫印迹法检测血小板源性生长因子B(PDGFB)、血小板源性生长因子受体β(PDGFRβ)、磷酸化PDGFRβ(p-PDGFRβ)、磷脂酰肌醇3激酶(PI3K)、蛋白激酶B(Akt)和磷酸化Akt(p-Akt)蛋白表达水平。结果高、低剂量组细胞增殖率、迁移能力、细胞侵袭数低于对照组,且高剂量组低于低剂量组(P<0.05)。高、低剂量组PDGFB、PDGFRβ、p-PDGFRβ、PI3K、p-Akt蛋白表达低于对照组,且高剂量组低于低剂量组(P<0.05)。3组Akt蛋白表达比较差异无统计学意义(P>0.05)。结论薏苡仁提取物可抑制人OSCC CAL27细胞增殖、迁移和侵袭能力,作用机制可能与抑制PDGFB/PDGFRβ/PI3K/Akt轴活性有关,且干预浓度越高抗肿瘤效果越佳,为薏苡仁提取物对OSCC的分子靶向治疗提供新的理论依据。 Objective To investigate effects of different concentrations of Coix Seed extract on proliferation,migration and invasion of oral squamous cell carcinoma(OSCC)cells and its mechanism.Methods Human OSCC CAL27 cells were cultured in vitro and divided into high-dose group,low-dose group and control group according to the experimental design.The high-dose and low-dose groups were intervened with 20μmol/L and 10μmol/L Coix Seed extract respectively,while control group was not given any intervention.The cells in each group were cultured for 24 h,and then MTT method,cell scratch method and Transwell method were used to detect the cell proliferation rate,migration ability and in vitro invasion ability respectively.In the meantime,western blot method was used to detect protein expressions platelet-derived growth factor B(PDGFB),platelet-derived growth factor receptorβ(PDGFRβ),phosphorylated PDGFRβ(p-PDGFRβ),phosphatidylinositol 3 kinase(PI3K),protein kinase B(Akt)and phosphorylated Akt(p-Akt).Results Values of cell proliferation rate,migration ability and cell invasion number in high-dose and low-dose groups were significantly lower than those in control group,and the values in high-dose group were significantly lower than those in low-dose group(P<0.05).Protein expressions of PDGFB,PDGFRβ,p-PDGFRβ,PI3K and p-Akt in high-and low-dose groups were significantly lower than those in control group,and the protein expressions in high-dose group were significantly lower than those in low-dose group(P<0.05).There were no significant differences in Akt protein expression among three groups(P>0.05).Conclusion Coix Seed extract may inhibit the proliferation,migration and invasion of human OSCC CAL27 cells,and the mechanism may be related to the inhibition of PDGFB/PDGFRβ/PI3K/Akt axis activity,and the higher the intervention concentration is,the better the anti-tumor effect is.This provides a new theoretical basis for molecular targeted therapy of OSCC by Coix Seed extract.
