摘要
目的:采用高效液相-四级杆-飞行时间质谱(HPLC-Q-TOF-MS)技术定性分析麻黄附子细辛汤化学成分,以及采用HPLC-VWD法定量分析麻黄附子细辛汤中的6种成分。方法:采用HPLC-Q-TOF-MS技术,使用PeakView软件、参考相关标准品及文献对麻黄附子细辛汤中的化学成分进行鉴定分析。同时,采用HPLC-VWD法测定麻黄附子细辛汤中麻黄碱、伪麻黄碱、苯甲酰乌头原碱、苯甲酰新乌头原碱、苯甲酰次乌头原碱及细辛脂素的含量。结果:该方中共鉴定出39个化合物,包括附子中27个化合物、麻黄中11个化合物,细辛中2个化合物(其中1个与麻黄共有)。定量分析的6个成分在各自范围内线性关系较好(r>0.9993);加样回收率98.99%~101.53%;RSD0.74%~1.19%。结论:上述方法灵敏度高、准确性好,可用于麻黄附子细辛汤的定量及定性检测。
OBJECTIVE To establish HPLC-Q-TOF-MS for rapid identification of constituents from Mahuang Fuzi Xixin Decoction,and HPLC-VWD methods was used to determination of the quantitatively analyze the six components of Mahuang Fuzi Xixin Decoction.METHODS The chemical constituents in Mahuang Fuzi Xixin Decoction were identified and analyzed by HPLC-Q-TOF-MS,peakview software,relevant standards and literature HPLC-VWD was used to simultaneous determination of six constituents,including ephedrine,pseudoephedrine,benzoylmesaconine,benzoylaconine,benzoylhypacoitine,asarinin.RESULTS Thirty-nine constituents were identified in Mahuang Fuzi Xixin Decoction,27 compounds from aconite,11 compounds from ephedra and 1 compound from Asarum(common with ephedra).Six constituents showed good linear relationships within their own ranges(r>0.9993),whose average recoveries were 98.99%-101.53%with the RSDs of 0.74%-1.19%.CONCLUSION This method is stable and reliable,and can be used for the quality control of Mahuang Fuzi Xixin Decoction.
