摘要
[目的]探讨HOXC9在肝细胞癌(HCC)组织中的表达以及对癌细胞增殖、迁移和侵袭的影响。[方法]收集2015~2019年于我院接受手术治疗的65例HCC患者的癌组织及相应癌旁组织,通过免疫组化检测HOXC9在癌组织及癌旁组织中的表达情况,通过RT-qPCR检测HOXC9在肝上皮细胞THLE-3、HCC细胞系HepG2、SMMC-7721、Huh7细胞中HOXC9 mRNA的表达,选取HepG2细胞用于后续实验。采用脂质体2000将siRNA-NC以及siRNA-HOXC9转染HepG2细胞,作为si-NC组、si-HOXC9组,对照组细胞不处理。通过RT-qPCR检测转染后各组细胞HOXC9 mRNA的表达,通过MTT检测各组细胞增殖情况,Transwell和划痕实验检测侵袭和迁移能力,Western-blot检测各组细胞Ki-67、CyclinD1、MMP-2、MMP-9蛋白表达水平。[结果]HOXC9在癌组织中阳性表达率高于癌旁组织(P<0.05)。与肝上皮细胞THLE-3比,HOXC9在HCC细胞系中表达升高,干扰HOXC9后,HepG2细胞增殖、迁移和侵袭能力降低,Ki-67、CyclinD1、MMP-2、MMP-9蛋白表达水平均下降(P<0.05)。[结论]HOXC9在HCC组织及HCC细胞系中高表达,干扰HOXC9能够抑制HCC细胞增殖、迁移和侵袭。
[Objective]To investigate the expression of HOXC9 in hepatocellular carcinoma(HCC)tissues and its role on the proliferation,migration and invasion of hepatocellular carcinoma cells.[Methods]The tumor tissues and adjacent tissues from 65 HCC patients who received surgical treatment in our hospital from 2015 to 2019 were collected,and the expression of HOXC9 in HCC tissues and adjacent tissues was detected by immunohistochemical.The expression of HOXC9 mRNA in THLE-3 liver epithelial cells,hepatoma cell line HepG2,SMMC-7721 and Huh7 cells was detected by RT-qPCR,and HepG2 cells were selected for subsequent experiments.HepG2 cells were transfected with siRNA-NC and siRNA-HOXC9 using Lipofectamine 2000,and were treated as si-NC group and si-HOXC9 group,while the control group cells were not treated.HOXC9 mRNA expression in the transfected cells was detected by RT-qPCR,cell proliferation was detected by MTT,invasion and migration ability were detected by Transwell and scratch assay,and protein expression levels of Ki-67,CyclinD1,MMP-2 and MMP-9 were detected by Western-blot.[Results]The positive expression rate of HOXC9 in HCC tissues was higher than that in adjacent tissues(P<0.05).Compared with THLE-3 ratio of liver epithelial cells,HOXC9 expression was increased in liver cancer cell lines.After HOXC9 interference,HepG2 cell proliferation,migration and invasion ability were decreased,and protein expression levels of Ki-67,CyclinD1,MMP-2 and MMP-9 were all decreased(P<0.05).[Conclusion]HOXC9 is highly expressed in hepatocellular carcinoma tissues and hepatocellular carcinoma cell lines.Interference with HOXC9 can inhibit the proliferation,migration and invasion of liver cancer cells.
