摘要
目的分离鉴定药用植物钩藤的化学成分。方法钩藤茎枝用90%乙醇回流提取,通过硅胶柱色谱、高效液相色谱、中低压液相色谱、葡聚糖凝胶柱色谱、制备薄层色谱等多种色谱分离技术对乙醇提取物分离和纯化;采用高分辨质谱及核磁共振波谱法确定化合物结构。结果从钩藤中分离鉴定了11个化合物,分别为高丽槐素(1)、去氢钩藤碱(2)、3,4,5,-三甲氧基苯酚(3)、喜树次碱(4)、(-)-N-甲基金雀花碱(5)、毛钩藤碱(6)、去氢毛钩藤碱(7)、缝籽嗪甲醚(8)、3α-二氢卡丹宾碱(9)、卡丹宾碱(10)和喜果苷(11)。结论从钩藤的茎枝中分离鉴定了9个生物碱类化合物,1个黄酮类化合物和1个酚类化合物,其中化合物1,4和5首次从该属植物中分离得到,为钩藤的进一步开发利用提供参考。
Purposes—To study the chemical constituents of Uncaria rhynchophylla(Miq.)Miq.ex Havil..Methods—The stems of U.rhynchophylla were extracted by 90%ethanol.Separation and purification of the chemical constituents were carried out by silica gel column chromatography,HPLC,MPLC,sephadex gel chromatography and preparative thin layer chromatography.Their structures were identified by high resolution electrospray ionization mass spectroscopy(HR-ESI-MS)and NMR.Result—Eleven compounds were identified from U.rhynchophylla,namely,maackiain(1),corynoxeine(2),3,4,5-trimethoxyphenol(3),venoterpine(4),(-)-N-methylcytisine(5),hirsutine(6),hirsuteine(7),geissoschizine methylether(8),3α-dihydrocadambine(9),cadambine(10),and vincosamide(11).Conclusion—Nine alkaloids,one flavonoid and one phenolic compound were isolated and identified from U.rhynchophylla.Compounds 1,4 and 5 were isolated from this genus for the first time,which provided references for the further exploitation and utilization of U.rhynchophylla resources.
