高效液相色谱法测定尿素的含量

摘要 目的建立高效液相色谱法测定尿素的含量。方法采用Waters Atlantis T3色谱柱(4.6 mm×250 mm,5μm),流动相为水,流量0.9 ml/min,检测波长195 nm,柱温30℃,进样体积10μl。结果在0.1074~0.5368 mg/ml的范围内线性关系良好(r=0.999,n=5);平均回收率为99.42%,其RSD=0.8%。结论本方法简便、准确,重复性好,专属性强,灵敏度高,可用于尿素的含量测定。 Objective To establish a HPLC method for determination of Urea.Methods The determination was performed by using Waters Atlantis T3 column(4.6 mm×250 mm,5μm)with the mobile phase of water.The flow rate was 0.9 ml/min,and the detection wavelength was set at 195 nm.The column temperature was 30℃,and the volume of injection was 10μl.Results The linearity range was good in the range of 0.1074~0.5368 mg/ml(r=0.999,n=5).The average recovery rate was 99.42% with RSD as 0.8%.Conclusion The method is simple,and has the good accuracy,repeatability,specificity and sensitivity.It can be applied to the determination of Urea.
