钛表面PDLLA-LN5复合涂层的构建及对角质形成细胞生物学行为的影响 被引量:1

摘要 目的:在纯钛表面构建聚消旋乳酸-层粘连蛋白5(PDLLA-LN5)复合缓释涂层,观察对角质形成细胞生物学行为的影响。方法:以纯钛为基底(P组),表面滴加PDLLA(PDLLA组),通过层层组装-物理吸附的方法制备载LN5的PDLLA涂层(PDLLA/LN5组),扫描电镜观察试样表面形貌,ELISA法测定LN5的释放曲线。将HaCaT细胞分别培养在3组试样表面,扫描电镜观察接种后细胞形态;CCK8法测定细胞增殖;DAPI染色后共聚焦显微镜观察接种后细胞黏附数量;细胞划痕法分析细胞迁移情况。结果:PDLLA-LN5复合涂层可以实现28 d以上LN5的持续释放;PDLLA/LN5组较P组和PDLLA组显著促进角质形成细胞增殖与黏附(P<0.05);与PDLLA组相比,PDLLA/LN5组划痕愈合百分比更高(P<0.05)。结论:钛表面PDLLA-LN5涂层对角质形成细胞的增殖、黏附及迁移具有促进作用。 Objective:To fabricate a poly(D,L-lactide)(PDLLA)controlled-release coating loading laminin-5(LN5)on titanium surface and to explore its effects on biological behavior of keratinocyte.Methods:Pure titanium was used as the base(group P).PDLLA was prepared on titanium surface(group PDLLA),then LN5 was loaded on PDLLA surface by layer-by-layer deposition and physical adsorption(group PDLLA/LN5).The morphology of PDLLA-LN5 coating was observed by SEM.The concentration of LN5 released by the PDLLA-LN5 composite coating was tested by ELISA.HaCaT cells were respectively cultured on the surfaces of the specimens of the 3 groups.SEM was used to observe the morphology of the cells,and the cell proliferation was assessed by CCK8 assay.After fluorescent staining by DAPI,confocal microscope was used to count the number of cell adhesion after seeding.Wound-healing scratch assay was used to measure cell migration after scratching.Results:The PDLLA-LN5 coating showed sustained release of LN5 that lasted for at least 28 days.Compared with P and PDLLA,PDLLA/LN5 promoted the proliferation and adhesion of HaCaT cells(P<0.05).Compared with the PDLLA group,the PDLLA/LN5 group showed a higher percentage of scratch healing(P<0.05).Conclusion:The PDLLA-LN5 coating on titanium can improve the proliferation,adhesion and migration of HaCaT cells.
