中华眼镜蛇咬伤导致局部组织坏死的猪模型构建 被引量:2

摘要 目的通过广西巴马小香猪建立中华眼镜蛇咬伤导致局部组织坏死的动物模型。方法选择健康的广西巴马香猪12头,分为蛇毒组和对照组各6头,每头猪按照本课题组前期获得的半数致死量为剂量依据,蛇毒组以浓度5 mg/kg,剂量0.2 ml/kg于实验猪右后侧大腿肌肉注射,对照组注射等量生理盐水。注射蛇毒后观察实验猪的生物学行为和局部伤口变化情况,采用苏木精-伊红染色(Hematoxylin-Eosin staining,HE)观察注射部位肌肉组织的病理学变化,并进行血液学分析。结果蛇毒组实验猪注射中华眼镜蛇毒3 h后,注射部位伤口开始出现淤青肿胀,随后肿胀部位不断扩大;6~12 h,出现渗出液外渗,且伴有出血;24 h,伤口出现糜烂性溃疡,且缓慢向四周扩散;第3天时,伤口部位皮肤向下凹陷,出现坏死;第6天,出现伤口全部坏死溃烂;第7天,坏死部分全部脱落。对照组实验猪注射部位全程无变化。同时蛇毒组在注射中华眼镜蛇毒后6 h,实验猪腿部肌肉肌丝的排列出现紊乱,没有规则;注射后12 h,肌肉组织中细胞溶解,见浆液性渗出物弥漫浸润组织,局部出现炎性水肿细胞质呈现不规则排布,细胞核成团聚集在组织间浆液中;24 h后,肌肉组织细胞溶解加重,出现坏死,部分细胞核凋亡消失;在36 h,仅剩少许肌肉组织,成形细胞核完全消失;48 h后,肌肉组织全部溶解,形成空泡状,说明注射部位组织全部溃烂坏死。对照组实验猪腿部肌肉肌丝排列整齐,肌肉组织细胞形态完好,清晰可见细胞核全程无较大变化。与对照组各时间点比较,蛇毒组实验猪的WBC、RBC、LDH和CK水平均显著升高,而且AST与ALT在24 h内急剧上升,组间比较差异均具有显著统计学意义(均P<0.05)。结论成功建立中华眼镜蛇毒致局部组织坏死的猪模型,为蛇伤的救治提供良好实验模型。 Objective To establish a Guangxi Bama minipig model for local tissue necrosis on Naja naja atra bite.Methods Twelve healthy Guangxi Bama fragrant pigs were selected and divided into 6 pigs in the snake venom group and 6 pigs in the control group,and each pig was injected intramuscularly with a concentration of 5 mg/kg and a dose of 0.2 ml/kg in the right hind side of the thigh of the experimental pigs according to the half lethal dose obtained by our group in the early stage.The biological behavior and local wound changes of the experimental pigs were observed after the injection of snake venom,and the pathological changes of muscle tissue at the injection site were observed by Hematoxylin-Eosin staining(HE),and hematological analysis was performed.Results 3 h after the injection of Naja naja atra venom,the wound at the injection site began to appear bruised and swollen,and then the swollen area expanded continuously;6-12 h later,exudate appeared with bleeding;24 h later,vesicular ulcers appeared in the wound and spread slowly in all directions;on the third day,the skin at the wound site sunk downward and necrosis appeared;on the sixth day,all necrotic ulcers appeared in the wound;on the seventh day,all necrotic parts of the wound fell off.On the 7th day,all necrotic parts fell off.In the control group,there was no change at the injection site throughout.At the same time,6 h after the injection of Naja naja atra venom,the arrangement of muscle filaments in the leg of the experimental pigs appeared disorganized and irregular;12 h after the injection,the cells in the muscle tissue were lysed,and the plasma exudate was seen to diffuse and infiltrate the tissue,and the local inflammatory edema cytoplasm showed irregular arrangement,and the nuclei gathered in the intertissue plasma in clusters;24 h later,the cell lysis in the muscle tissue was aggravated and necrosis appeared,and some of the At 36 h,only a little muscle tissue remained,and the nuclei of formed cells completely disappeared;after 48 h,all muscle tissues
