摘要
本研究以海南省青葛港马尾藻属(Sargassum spp.)为研究对象,利用coxⅠ(细胞色素C氧化酶亚基I)、ITS(内转录间隔区)序列及rbcL(1,5-二磷酸核酮糖羧化酶/加氧酶大亚基)3种DNA条形码分析马尾藻属遗传进化关系。结果表明,(1)基于形态鉴定及利用coxⅠ、ITS和rbcL三种DNA条形码进行分子鉴定的方式鉴定出9种海南省青葛港常见马尾藻属物种;(2)利用rbc L基因扩增出DNA序列的GC含量最低(32%),ITS基因扩增的DNA序列GC含量最高(59%),且在不同样本间ITS扩增的DNA序列GC含量差异最大,在57%~59%之间;(3)coxⅠ条形码序列可聚为两类,其中6个几乎不存在遗传距离,且5个样本间的遗传距离均小于50;从rbcL条形码序列的聚类结果看来,除了RQL3之外,其他样品聚为一类,6个样本间的遗传距离均小于50。ITS条形码序列聚类的结果显示,9种马尾藻样本可聚为两类,马尾藻样本之间均存在一定的遗传距离。研究认为,青葛港海域马尾藻种内变异丰富,同一属的样品表现出很好的属内种间多样性;在分析青葛港马尾藻的遗传进化时利用ITS基因比其他两种基因能发现更多的遗传变异,能有效分析马尾藻的遗传进化关系。该研究结果对研究马尾藻种群生长和遗传进化具有重要现实意义,可为人工培育优异马尾藻属及其选育打下基础。
Using the coxⅠ(cytochromec oxidase subunit I gene),ITS(internal transcribed spacer)and rbcL(Ribulose-1,5-bisphosphate carboxylase/oxygenase)to analyze the genetic evolutionary relationships among 9 species of Sargassum spp.of Qingge Harbor,Hainan province.The results showed that:(1)Based on morphological identification and molecular identification with three DNA barcodes,9 species of Sargassum were identified in Qingge Harbor,Hainan Province.(2)The GC content of the DNA sequence amplified by rbcL barcode was the lowest(32%),by ITS barcode was the highest(59%);and ITS barcode varied the most among different samples,ranging from 57%to 59%.(3)Cox I barcode sequences can be grouped into two categories,six of which have almost no genetic distance,and the genetic distance between the five samples is less than 50.From the clustering results of rbcL barcode se quences,it can be seen that the other samples are clustered into one category except RQL3,the genetic distance between 6 samples was less than 50.The results of ITS barcode sequence clustering showed that 9 samples of Sargassum could be classified into two categories,and there were some genetic distance between the samples.The results showed that there were abundant intraspecific variations of Sargassum in Qingge Harbor area,and the samples of the same genus showed good intra-genus and inter-species diversity.In the analysis of the genetic evolution of Sargassum Qingge Harbor area,the use of ITS gene than the other two genes can find more genetic variation,can effectively analyze the genetic evolution of Sargassum.The results of this study have important practical significance for the study of population growth and genetic evolution of Sargassum,and lay a foundation for the artificial breeding of Sargassum.
