摘要
目的:以白假丝酵母菌感染雌激素化的小鼠形成阴道炎模型,并探讨美廉净凝胶对白假丝酵母菌性阴道炎的药效作用。方法:建立白假丝酵母菌感染小鼠阴道炎模型,设置模型组,双唑泰组,保妇康凝胶组,美廉净凝胶高、中、低剂量组,阴道给药每天2次,连续14 d。测定小鼠血清中白细胞介素-6(interleukin-6,IL-6)、NO(Nitric Oxide)的表达水平,并以其阴道病原体转阴率和阴道组织炎症治愈程度作为评价指标。结果:与模型组比较,随治疗时间的延长,美廉净凝胶能在一定程度上减轻白假丝酵母菌小鼠阴道炎的水肿,抑制分泌物中白假丝酵母菌增长,减少菌落的数量,炎症评分显著性下降(P<0.01),但对减少分泌物量的作用不明显;脏器指数中,美廉净凝胶对肝脏指数和阴道指数影响不大,但能提高脾脏指数和胸腺指数;各受试药物组血清中IL-6和NO的含量相对模型组均有所降低(P<0.01),美廉净凝胶各剂量组抑制IL-6的作用对与阳性药相当;HE染色观察到美廉净凝胶能减轻阴道炎症,PAS染色可观察到美廉净凝胶能减轻白假丝酵母菌的侵染和糖原减少。结论:美廉净凝胶能通过抑制阴道内白假丝酵母菌的增殖,降低血清中IL-6和NO的含量来减轻小鼠的阴道感染。
OBJECTIVE To evaluate the curative effects of Meilianjing temperature-sensitive gel on a mouse model of vaginitis induced by C. albicans.METHODS The mouse model of vaginitis was induced by C. albicans, and then divided into model group, Shuangzuotai group, Baofukang gel group, and low, medium and high dose of Meilianjing gel groups. Except those in the model group, the mice in the other groups were treated with corresponding drug twice a day for 14 consecutive days. After treatment, the expression levels of interleukin-6(IL-6) and NO in serum of mice were measured. Furthermore, the vaginal pathogen conversion rate and vaginal tissue inflammation degree were analyzed to determine the curative effects of Meilianjing gel.RESULTS Compared with the model group, Meilianjing gel could inhibit the growth and reduce the number of colonies of C. albicans in mouse vagina. The edema and inflammation of mouse vagina were also attenuated after treatment, while Meilianjing gel showed little effect of secretion inhibition. Meanwhile, Meilianjing gel had little effect on the liver index and vaginal index, but it could increase the spleen and thymus index. The contents of IL-6 and NO in serum in every drug treatment group were lower than those in the model group(P<0.01). HE staining showed that Meilianjing gel could reduce vaginitis, and PAS staining showed that it could reduce the infection and glycogen reduction of C. albicans.CONCLUSION Meilianjing gel can attenuate the vaginitis by inhibiting the proliferation of C. albicans and reducing the expression of IL-6 and NO in serum of mice.
