摘要
采用陶瓷膜并通过酒精透析方式提取了亲脂性化合物利普斯他汀。根据陶瓷膜通量和乙醇耗量确定了利普斯他汀提取的基本参数:陶瓷膜孔径为50 nm、膜面流速为5 m/s、平均跨膜压力为0.10 MPa,发酵液浓缩倍数达到2倍时开始流加95%乙醇,乙醇流加速度和渗透液通量一致,维持循环罐液位不变,渗透液酒精浓度达到55度时开始收集渗透液,95%乙醇总用量为2倍体积,此时,利普斯他汀回收率达到95.0%,满足了生产需求。采用控制跨膜压力的方法显著提高了利普斯他汀渗透液通量,并为进一步提高陶瓷膜设备的渗透液通量及扩大适用范围提供了思路。
Ceramic membrane technology is applied in the ethanol dialysis of lipophilic lipstatin from fermentation broth.Based on permeation flux of ceramic membrane and ethanol consumption,the basic parameters and methods for extraction of lipstatin are determined as follows:the pore size of ceramic membrane is 50 nm,membrane surface velocity is 5 m·s^(-1),the average transmembrane pressure is 0.1 MPa,95%ethanol begins to feed continuously when the fermentation broth is concentrated to 2 times,the feeding speed of ethanol is consistent with the permeation flux to maintain a constant level in the circulating fermentation tank,the collection of permeation broth is started when the content of ethanol in permeation broth reaches 55%,total ethanol consumption are 2 times.After these operations,the recovery yield of lipstatin reaches 95%,which can meet the production specifications.The technique to control the average transmembrane pressure can significantly enhance the permeation flux of lipstatin extraction,which provides ideas for further enhancing the permeation flux of ceramic membrane and expanding its application scope.
