摘要
目的通过生物信息学方法,筛选可能与结肠癌诊断、预后及免疫浸润相关的基因。方法从GEO数据库中下载数据集,并筛选出结肠癌差异基因,通过STRING数据库和Cytoscape软件筛选出核心基因CXC型趋化因子配体2(CXC chemokine ligand 2,CXCL2)和血小板反应蛋白2(thrombospondin-2,THBS2)。通过GEPIA和UALCAN数据库分析CXCL2和THBS2表达水平与预后关系以及其表达与结肠癌患者临床特征之间的关系。TIMER数据库分析CXCL2和THBS2与结肠肿瘤免疫细胞浸润的相关性。LinkedOmics和DAVID数据库分析CXCL2和THBS2基因的正相关基因和KEGG通路。结果鉴定出结肠癌相关的核心基因16个,包括CXCL2和THBS2。CXCL2和THBS2在结肠癌中的表达均高于正常组织(均P<0.05),且在年龄、性别、分期和淋巴结转移方面比较,差异均具有统计学意义(均P<0.05)。CXCL2高表达组患者5年生存率高于低表达组(P<0.05),THBS2高表达组患者5年生存率低于低表达组(P<0.05)。CXCL2和THBS2基因参与结肠癌肿瘤细胞的免疫浸润。KEGG通路显示CXCL2主要参与细胞因子受体的相互作用、肿瘤坏死因子(tumor necrosis factor,TNF)信号通路和趋化因子信号通路等。THBS2主要参与细胞外基质(extracellular matrix,ECM)-受体相互作用、血小板活化和磷脂酰肌醇3-激酶/蛋白激酶B(phosphatidylinositide 3-kinases/protein kinase B,PI3K-Akt)信号通路等。结论CXCL2和THBS2为结肠癌诊断及预后相关基因,参与多种分子调控通路,可作为结肠癌潜在的生物标志物。
Objective To screen genes related to the diagnosis,prognosis and immune infiltration of colon cancer through bioinformatics analysis.Methods The data set was downloaded from the GEO database and the differential genes of colon cancer were screened.The core genes CXC chemokine ligand 2(CXCL2)and thrombospondin-2(THBS2)were screened through STRING database and Cytoscape software.The relationship between the expression of CXCL2 and THBS2 and the prognosis and the relationship between the expression and the clinical characteristics of colon cancer patients were analyzed through the GEPIA and UALCAN databases.TIMER database was used to analyze the correlation between CXCL2 and THBS2 and immune cell infiltration of colon tumors.LinkedOmics and DAVID database were used to analyze the positively related genes of CXCL2 and THBS2 and the KEGG pathway.Results Sixteen core genes related to colon cancer were identified,including CXCL2 and THBS2.The expression of CXCL2 and THBS2 in colon cancer were both significantly higher than those in normal tissues(both P<0.05),and the differences in age,sex,stage,and lymph node metastasis were all statistically significant(all P<0.05).The 5-year survival rate of patients with high expression of CXCL2 was higher than that of patients with low expression of CXCL2(P<0.05),and the 5-year survival rate of patients with high expression of THBS2 was lower than that of patients with low expression of THBS2(P<0.05).CXCL2 and THBS2 genes participated in the immune infiltration of colon cancer cells.KEGG pathway showed that CXCL2 was mainly involved in cytokine receptor interaction,tumor necrosis factor(TNF)signaling pathway,and chemokine signaling pathway.THBS2 was mainly involved in the interaction with extracellular matrix(ECM)-receptor,platelet activation,and phosphatidylinositide 3-kinases/protein kinase B(PI3K-Akt)signaling pathway.Conclusions CXCL2 and THBS2,as genes related to the diagnosis and prognosis of colon cancer,are involved in a variety of molecular regulatory pathways,and can be u
