摘要
目前研究显示代谢综合征(MetS)患者发生认知功能损伤的风险增高,且针对MetS各异常代谢组分的治疗被认为可预防或延迟痴呆的发病。磷脂酰肌醇-3激酶/蛋白激酶B(PI3K/Akt)信号通路在外周代谢和脑功能的正常运行中起着重要作用,是MetS与认知功能损伤发生机制中共同存在的信号通路,目前已有研究尝试以此信号通路作为治疗MetS相关认知功能损伤的切入点。本文从脑对葡萄糖的摄取、脑内胆固醇合成、脑内乙酰胆碱水平、脑内β-淀粉样蛋白聚集和tau蛋白磷酸化等几个方面,对MetS通过PI3K/Akt信号通路损伤认知功能的机制研究进展进行综述,旨在理清PI3K/Akt信号通路是如何参与MetS损伤认知功能的,以期寻找到治疗MetS相关认知功能损伤的新靶点。
Multiple studies has indicated an increased risk of cognitive decline in patients with metabolic syndrome(MetS),and treatments targeting the abnormal metabolic components of MetS are considered to be able to prevent and delay the onset of dementia.Phosphatidylinosito-3 kinase/protein kinase B(PI3K/Akt)signaling pathway plays an important role in peripheral metabolism and brain biological function,and is a common signaling pathway in the pathogenesis of MetS and cognitive impairment.PI3K/Akt signaling pathway has been attempted to use as an entry point for treatment of cognitive impairment related with MetS.In our research,the mechanism of cognitive impairment in MetS through PI3K/Akt signaling pathway is reviewed from aspects of glucose uptake,cholesterol synthesis,acetylcholine level,Aβaggregation and tau protein phosphorylation in brain,which aims to clarify how PI3K/Akt signaling pathway participates in cognitive impairment related to comorbid MetS,and to find new targets for treating cognitive impairment related to comorbid MetS.
