摘要
目的:检测宫颈鳞状细胞癌组织和细胞中3-磷酸甘油酸脱氢酶(3-phosphoglycerate dehydrogenase,PHGDH)的表达水平,探讨其在肿瘤发生发展中的作用。方法:使用实时荧光定量PCR法和免疫组织化学法检测宫颈鳞状细胞癌和正常宫颈组织中PHGDH mRNA转录水平和蛋白翻译水平;利用实时荧光定量PCR法和Western blot法检测HeLa、Siha和Hacat细胞系中PHGDH mRNA转录水平和蛋白翻译水平。结果:与正常宫颈组织比较,宫颈鳞状细胞癌组织中PHGDH mRNA转录水平升高,PHGDH蛋白表达水平升高,其差异具有统计学意义(P<0.05);HeLa和Siha细胞系中PHGDH mRNA转录水平较Hacat细胞系高,PHGDH蛋白表达水平也较Hacat细胞系高,其差异具有统计学意义(P<0.05)。结论:宫颈鳞状细胞癌组织和细胞系中PHGDH高表达,提示PHGDH可能与促进宫颈鳞状细胞癌病变进展有关。
Objective:To detect 3-phosphoglycerate dehydrogenase(PHGDH)expression in cervical squamous cell carcinoma tissues and cells and its role in tumor development.Methods:Real-time fluorescence quantitative PCR and immunohistochemistry were used to detect the levels of PHGDH mRNA transcription and protein translation in cervical squamous cell carcinoma tissues and normal cervical tissues.Real-time fluorescence quantitative PCR and Western blot were used to detect the levels of PHGDH mRNA transcription and protein translation in HeLa,Siha and Hacat cell lines.Results:The PHGDH mRNA transcription level and PHGDH protein expression level in cervical squamous cell carcinoma tissues were significantly higher than that in normal cervical tissues(P<0.05),and the PHGDH mRNA transcription level and PHGDH protein expression level in HeLa and Siha cells were higher than that in Hacat cells.The difference was statistically significant(P<0.05).Conclusion:The high expression of PHGDH in cervical squamous cell carcinoma tissues and cell lines suggests that PHGDH may be related to cervical squamous cell carcinoma progression.
