黄粉虫粗多糖的初步纯化及体外抗氧化活性研究 被引量:1

Preliminary Purification and in Vitro Antioxidant Activity of Crude Polysaccharides from Tenebrio Molitor

摘要 以黄粉虫粗多糖为原料,通过单因素脱蛋白试验,利用响应面法优化脱蛋白工艺,经DEAE-纤维素阴离子层析柱纯化,对多糖进行体外抗氧化活性分析。结果表明:各因素对黄粉虫粗多糖蛋白去除率的影响程度为:脱蛋白次数>振摇时间>静置时间。当Sevage试剂加入量与粗多糖溶液体积比为1:1时,优化条件为:振摇时间17 min、静置时间19 min、脱蛋白次数2次,蛋白去除率达61.25%。与未除蛋白的粗多糖相比,除蛋白后黄粉虫多糖对DPPH自由基、羟自由基清除能力的IC_(50)减小,分别为3.052、7.276 mg/mL,说明多糖抗氧化活性增强。黄粉虫多糖经不同浓度NaCl溶液梯度洗脱纯化获得4个主要组分,其中0.5 mol/L NaCl洗脱的多糖体外抗氧化性综合能力最强。 The crude polysaccharide from Tenbrio molitor was used as the raw material,the deproteinization process was optimized by single factor test and the response surface methodology,then purified by DEAE-celluiose anion column,and the antioxidant activity of polysaccharides was analyzed in vitro.The results showed that the effects of various factors on the protein removal rate of Tenbrio molitor pleysaccharides were as follows:deproteinization times>shaking time>resting time.When the ratio of Sevage reagent to polysaccharide solution was 1:1,shaking time was 17 min,resting time was 19 min,and deproteinzation times were 2 times,the removal rate of protein was 61.25%.Compared with the crude polysaccharides without removing protein,the IC;value of DPPH radical and OH radical scavenging rate of polysaccharides after removing protein decreased to 3.052 mg/mL,7.276 mg/mL,that suggest the antioxidant activity was enhanced.Four major components of Tenbrio molitor polysaccharides was obtained after different concentration of NaCl gradient elution purification,of which 0.5 mol/L NaCl eluant had the strongest antioxidant activity in vitro.
